peut réaliser au laboratoire les déterminations préliminaires
de la façon suivante :
Sédation par administration intra-péritonéale chez la souris Mesure de l'activité locomotrice. On place un groupe
de 3 souris dans un cylindre rond traversé par 6 faisceaux photoélectriques (Actophotomètro Métro). Un compteur enregistre chaque fois qu'une souris se déplace et intercepte un faisceau. La quantité qui détermine une dose à 50 % est colle qui produit une réduction d'activité de 50 % par rapport aux témoins recevant une solution saline, l'activité étant mosuréo sur 30 mn, en commençant 15 mn après l'injection.
Tranquillisation. Cet effet est mesuré par l'inhibition de la sensibilité conditionnée à une excitation chez les rats. Les tranquillisants ont pour effet de faire oublier aux rats de sauter sur un poteau en bois au son d'un bourdonnour pour éviter un choc électrique modéré provenant du sol métallique. La méthode est décrite par L. Cook et E. Weidlcy dans Annals of New York Academy of Sciences, vol. 66, page 740, 1957.
On a découvert qu'une bonne sédation (et tranquillisation) est obtenue quand l'unité XYN- dans la formule A est un groupe diméthylamino, pipérazino ou morpholino et 4-méthylpipéridino. Quand R3 dans la formule A est un alkyle, les 5 effets de sédation et de tranquillisation augmentent quand
l'alkyle augmente do dimension, du méthyle au butyle.
On peut mettre ces composés sous forme pharmaceutique usuelle, par exemple en comprimas, en capsules, on sirop, en suppositoires, etc., on suivant les techniques classiques en pharmacie, avec l'emploi dos véhicules et dos excipients appropriés.
