Méthode de préparation des composés de formule généraleI
On part d'un chlorure de p-alkylbenzyle (I) (obtenu par chlorométhylation d'un p-alkylbenzène), lequel est condensé avec un cyanure alcalin pour donner le p-alkylphénylacétonitrile corres- pondant (2);
ce dernier corps traité par l'acide sulfurique et un alcool (méthanol, éthanol ou propanbl) conduit au p-alkylphénylacé- tate correspondant (3) que l'on fait réagir à l'état brut sur l'hydroxylamine pour obtenir l'ecide phénylacéthydroxamique corres- pondant (4)

Préparation de l'acide n-tertbutylphénylacéthydroxamique

a) A une solution de 15,5 g de NaCN dans 15 cc d'eau, on ajoute en 30 minutes une solution de 45 g de chlorure de p-tertbutylbenzyle [point d'ébullition (3 mm/Hg): 88-89 ; obtenu par chlorométhylation

Le mélange ainsi obtenu est chauffé au reflux @@@ 4 heures, puis refroidi; le précipité inorganique est filtré lavé avec un peu d'alcool. L'alcool du filtrat est alor dis@@@@é, le résidu repris dans l'éther est lavé à l'eau et sécné sur du sulfate de magnésium. Après évaporation de l'éther, on distille sous pression réduite et on recueille le p-tertbutylphénylacétoni- trile, point d'ébullition (16 mm): 149-152). b) Mélanger sous agitation et en refroidissant 20 g (25 ml) de métha- nol et 20 g (10,9 ml) d'acide nulfurique concentré. Placer ensuite 17,3 g de p-tertbutylphënylacétonitrile et chauffer le tout à reflux pendant 10 heures. Refroidir et verser le mélange réactionnel aans un mélange eau-glace.
Séparer la couche organique, la sécher sur MgSOt et distiller le méthanol. On obtient ainsi à l'état brut le p-tertbutylphénylacétate de méthyle qu'or n'isole pas. b) Dissoudre à part 5 g de /la dans 150 ml de méthanol absolu et 7 g de MH20H-HC1 dans 100 ml de méthanol absolu. Mélanger les deux solu- tions et filtrer le précipité de NaCl formé.
Au filtrat obtenu, ajouter le p-tertbutylphénylacétate de méthyle brut et chauffer à reflux pendant 1 heure.
Après refroidissement, acidifier par HC1 à 20% et recueillir le pré- cipité d'acide p-tertbutylphénylacéthydroxamique formé.
Recristalliser dans l'acétone et recueillir le produit blanc d'un point de fusion de 115-118 . Le produit dissous dans le méthanol donne une coloration rouge-vin avec le chlorure ferrique (test des acides hydroxamiques).

Ce dernier réagit ensuite avec l'hydroxylamine pour donner l'acide hydroxamique (@) correspondant.

Dissoudre 7,2 g de Na dans 200 ml d'éthanol absolu, y ajouter 46 g de p-tertbutylphénol et ensuite 38 g de chloracétate d'éthyle.
Chauffer 1 heure à reflux ; le NaBr précipite dès le début de la réaction. Distiller alors l'alcool, diluer avec H20 et extraire 2 fois l'éther. Laver l'éther au carbonate à 10%, puis à l'eau, et sécher sur du sulfate de magnésium. Evaporer l'éther et distiller ensuite sous pression réduite.

Mélanger les deux solutions et filtrer le précipité de NaCl forme. Au filtrat obtenu, ajouter 23,6 g de l'ester ci-dessus et chauffer 1 heure à reflux.
Après refroidissement, acidifier par HC1 à 20% et recueillir le précipité formé. Recristalliser dans le benzène ; obtient ainsi l'acide p-tertbutylphénoxyacéthydroxamique d'un point de fusion de 149 - coloration rouge avec FeC13.

RESULTATS PHARMACOLO-UIQUES
La toxicité aiguë des divers composés étudiés s'est révélée faible, la LD 50 ne dépassant pas 1.200 à 2. 000 mg/kg chez la souris.
Diverses activités pharmacologiques ont pu être mises en évidence, parmi lesquelles il faut mentionner les pouvoirs anti- phlogistiques, analgésiques, tranquillisants et hypocholestérolé- miants.

Method of preparation of compounds of general formula I
One starts with a p-alkylbenzyl chloride (I) (obtained by chloromethylation of a p-alkylbenzene), which is condensed with an alkali cyanide to give the corresponding p-alkylphenylacetonitrile (2);
this last body treated with sulfuric acid and an alcohol (methanol, ethanol or propanbl) leads to the corresponding p-alkylphenylacetate (3) which is reacted in the crude state with hydroxylamine to obtain ecide corresponding phenylacethydroxamic (4)

The mixture thus obtained is heated to reflux for 4 hours, then cooled; the inorganic precipitate is filtered, washed with a little alcohol. Separate the organic layer, dry it over MgSOt and distill off the methanol. In this way, in the crude state, methyl p-tertbutylphenylacetate is obtained, which gold does not isolate. b) Separately dissolve 5 g of / la in 150 ml of absolute methanol and 7 g of MH20H-HCl in 100 ml of absolute methanol. Dissolve 7.2 g of Na in 200 ml of absolute ethanol, add 46 g of p-tertbutylphenol and then 38 g of ethyl chloroacetate.
Heat 1 hour at reflux; the NaBr precipitates from the start of the reaction. Then distil the alcohol, dilute with H20 and extract the ether twice. Wash the ether with 10% carbonate, then with water, and dry over magnesium sulfate. Evaporate the ether and then distill off under reduced pressure.

This gives ethyl p-tertbutylphenoxyacetate with a boiling point of 180-182 / 15 mm. b) Separately dissolve 5 g of Na in 150 ml of absolute methanol and 7 g of NH2OH.HCl in 100 ml of absolute methanol. Mix the two solutions and filter the precipitate from NaCl forms. To the filtrate obtained, add 23.6 g of the above ester and heat for 1 hour at reflux.
After cooling, acidify with 20% HCl and collect the precipitate formed. Recrystallize from benzene; thus obtains p-tertbutylphenoxyacethydroxamic acid with a melting point of 149 - red coloration with FeC13.

PHARMACOLO-UIC RESULTS
The acute toxicity of the various compounds studied was found to be low, the LD 50 not exceeding 1,200 to 2,000 mg / kg in mice.
Various pharmacological activities have been demonstrated, among which we must mention the anti-phlogiston, analgesic, tranquilizing and hypocholesterolemic powers.
As regards the analgesic and antiphlogistic powers, some of these hydroxamic acids, and among them p-isobutylphenylacethydroxamic acid, have shown an activity much greater than that of acetylsalicylic acid, in conventional tests:
