Nouveaux dérivés de la phénothiazine et leur préparation.
La présente invention a pour objet de nouveaux dérivés de la phénothiazine, leurs sels, leurs dérivés ammonium quaternaire et leurs procédés de préparation.

carboné divalent; n est égal à 0, ou 1. R1 et R2 représentent soit des atomes d'hydrogène ou des radicaux alcoyles inférieurs soit avec l'atome d'azote voisin des radicaux hétérocycliques monocycliques saturés tels que pyrrolidino, pipéridino, morpholino ou pipërazino éventuellement substitués.
Par radical inférieur dans le cas de X comme pour les autres symboles, il faut comprendre un radical renfermant au maximum 4 atomes de carbone.
Le radical hydrocarboné diraient A peut comporter de 2 à 4 atomes de carbone en chaîne droite ou ramifiée et tels que l'atome d'azote du noyau phénothiazine et celui du noyau pipéridine soient séparés par au moins deux atomes de carbone. Dans ces formules X, A, n, R1 et R2 ont les significations indiquées précédemment et Y représente un reste d'ester réactif tel qu'un atome d'halogène ou un reste d'ester sulfurique ou sulfonique.
Ce procédé, qui constitue le procédé préféré de l'invention, consiste à chauffer les réactifs à une température comprise entre 50 et 150 , de préférence dans un solvant organique inerte tel qu'un alcool ou un hydrocarbure aromatique par exemple benzène ou xylène, éventuelle- ment en présence d'un agent de condensation, jouant le rôle d'accep- teur d'acide, tel qu'un carbonate de métal alcalin; comme agent de condensation on peut également utiliser simplement un excès du com- posé pipéridinique.

Les nouveaux dérivés de la phénothiazine peuvent être transformés en sels d'addition avec les acides et en dérivés ammo- nium quaternaire.
Les sels d'addition peuvent être obtenus par action des nouveaux dérivés sur des acides dans des solvants appropriés. Comme solvants organiques on utilise par exemple des alcools, des éthers, des cétones ou des solvants chlorés; comme solvant minéral on utili- se avec avantage l'eau. Le sel formé précipite après concentration éventuelle de sa solution et est séparé par filtration ou décanta- tion.
Les dérivés ammonium quaternaire peuvent être obtenus par action des nouveaux dérivés sur des esters, éventuellement dans un solvant organique, à la température ordinaire ou plus rapidement par léger chauffage.
Les nouveaux dérivés de la phénothiazine de formule I peuvent être éventuellement purifiés par des méthodes physiques, telles que distillation, cristallisation, chromatographie, ou chimi- ques telles que formation des sels, cristallisation de ceux-ci et décomposition de ces derniers en milieu alcalin. Dans ces opérations, la nature de l'anion du sel est indifférente, la seule condition étant que le sel soit bien défini et aisément cristallisable.
Les nouveaux dérivés de la phénothiazine possèdent des propriétés pharmacodynamiques intéressantes; ce sont en particulier des dépresseurs du système nerveux extrêmement actifs, d'excellents

potentialisateurs de narcose, des tranquillisants, des antiémétiques puissants et de très bons analgésiques.
Pour l'emploi thérapeutique il est fait usage des nou- veaux composés soit à l'état de bases, soit à l'état de sels d'addi- tion ou dérivés ammonium quaternaire pharmaceutiquement acceptables, c'est-à-dire non toxiques aux doses d'utilisation. Les bases et sels d'addition définis ci-dessus constituent cependant la forme d'utili- sation préférée, c'est-à-dire celle dans laquelle les actions secon- daires indésirables sont les plus faibles et l'action principale la plus forte.
Comme exemples de sels d'addition pharmaceutiquement ac- ceptables peuvent être cités des sels d'acides minéraux (tels que les chlorhydrates, sulfates, nitrates, phosphates) ou organiques (tels que les acétates, propionates, succinates, benzoates, fumarates, maléates, oxalates, tartrates, méthanesulfonates, éthanedisulfonates, chlorothéophyllinates, théophyllineacétates, salicylates, phénol-

phtalinates, méthylène-bis-j3-oxynaphtoates) ou des dérivés de substi- tution de ces acides.
Comme exemples de dérivés ammonium quaternaire pharmaceu- @ tiquement acceptables peuvent être cités des dérivés d'acides miné- raux ou organiques tels que les chloro-, bromo- ou iodométhylates, -éthylates, -allylates ou -benzylates, les méthyl- ou éthyl-sulfates, les benzènesulfonates ou des dérivés de substitution de ces composés.
Les nouveaux composés peuvent être administrés à l'état pur ou en présence d'un diluant ou munis d'un enrobage. Les formes pharmaceutiques usuelles peuvent être utilisées, en particulier celles qui conviennent pour l'administration par voie orale, rectale ou parentérale. Les doses dépendent de l'effet thérapeutique recherché, de la voie d'administration, de la durée du traitement et de l'es- pèce animale. Elles sont généralement comprises entre 0,1 et 10 mg par kilogramme de poids .animal. En thérapeutique humaine on peut ad- mettre comme doses d'emploi journalières des doses de 20 mg à 600 mg de produit actif par voie orale et 10 mg à 500 mg par voie parenté- raie.

Les exemples suivants, donnés à titre non limitatif, montrent comment l'invention peut être mise en pratique.
EXEMPLE 1.-
On chauffe pendant 7 heures, à 2000, dans un autoclave,

une solution de 26 g de méthoxy-3(carbéthoxy-4" pipéridino)-3' propyl]-10 phénothiazine dans 100 cm3 d'éthanol avec 200 cm3 d'ammo- niac liquide.
Après refroidissement, on évapore l'ammoniac en tiédis- sant légèrement et on concentre ensuite à sec sous une pression de 15 mm de mercure environ.
On dissout le.résidu dans 200 cm3 d'acétate d'éthyle et on épuise la solution par 100 cm3 de solution d'acide méthanesulfonique 0,7 N; on filtre la phase aqueuse acide sur 1 g de noir et on l'al- calinise par 20 cm3 de solution de soude 4 N ; on extrait la base libérée avec de l'acétate d'éthyle, on sèche la solution avec du carbonate de potassium anhydre et on évapore à sec sous une pression de 15 mm de mercure environ.
Le résidu solide ainsi obtenu est finalement recristalli- sé dans l'acétonitrile; on obtient ainsi 12,5 g de méthoxy-3[(car- bamoyl-4" pipéridino)-3' propyl]-10 phénothiazine, poudre cristal- line blanc-crème fondant à 132-134 .

La méthoxy-3/*(carbéthoxy-4" pipéridino)-3' propy-17-10 phénothiazine de départ (dont l'oxalate acide fond à 130-132 ) est obtenue par condensation de la méthoxy-3 (chloro-3' propyl)-10 phénothiazine sur l'isonipécotate d'éthyle dans le xylène à reflux pendant 15 heures.
EXEMPLE 2. -
On chauffe pendant 24 heures à reflux une solution de 10 g de cyano-3 (chloro-3' propyl)-10 phénothiazine et 4,6 g d'isonipécotamide dans 100 cm3 d'éthanol, en présence de 3,5 g de carbonate de sodium anhydre; on rajoute ensuite 1,75 g de carbonate de sodium et chauffe à nouveau pendant 8 heures; après une nouvelle addition de le75 g de carbonate de sodium, on termine par un dernier

chauffage à reflux de 16 heures.
On chasse le solvant sous pression réduite (20 mm de mer- cure); on reprend le résidu par 50 cm3 d'eau et 150 cm3 d'acétate d'éthyle; après agitation, on sépare la phase organique et on l'é- puise avec 200 cm3 d'acide chlorhydrique N. On alcalinise la phase aqueuse avec 60 cm3 de solution de soude 4 N et on extrait la base libérée avec de l'acétate d'éthyle; on sèche la solution organique avec du sulfate de sodium et on concentre à sec sous pression réduite (20 mm de mercure).
Le résidu cristallisé jaune, pesant 10,5 g, est recristal- lisé successivement dans le benzène et dans l'éthanol. On obtient

La cyano-3 (chloro-3' propyl)-10 phénothiazine (P.F. = 139-140 ) est préparée par condensation du chloro-1 bromo-3 propane sur la cyano-3 phénothiazine dans l'ammoniac liquide en présence d'amidure de sodium-préparé "in situ".
L'isonipécotamide (P.F. 150-152 ) est préparée selon GROB et .al. Helv. 37, 1672 (1954).
EXEMPLE 3.-
On opère comme dans l'exemple n 2 à partir de 10 g de méthanesulfonyl-3 (chloro-3' propyl) -10 phénothiazine, 4 g d'iso- nipécotamide, 200 cm3 d'éthanol et 6 g au total de carbonate de so- dium anhydre.
Après recristallisation de la base cristallisée brute dans l'acétate d'éthyle, on obtient 6 g de méthanesulfonyl-3[(car-

samoy'-,-4" pipéridino)-3' propyl7-10 phénothiazine, poudre cristal- line jaune-pâle, fondant à 170-171 .
La méthanesulfonyl-3 (chloro-3' propyl)-10 phénothiazine (P. F. = 132 - 134 ) est obtenue par condensation du chloro-1 bromo-3 propane sur la méthanesulfonyl-3 phénothiazine dans l'ammo- niac liquide, en présence d'amidure de sodium préparé "in situ".

EXEMPLE 4.-
On opère comme dans l'exemple n 2 à partir de 10 g d'acétyl-3 (chloro-3' propyl)-10 phénothiazine, 4,45 g d'isonipéco- tamide, 200 cm3 d'éthanol et 7,4 g au total de carbonate de sodium - anhydre.
Après deux recristallisations successives de la base brute dans l'éthanol, on obtient 4,55 g d'acétyl-3 [(carbamoyl-4" pipéridino)-3' propyl]-10 phénothiazine, poudre cristalline jaune- orangé, P.F. : 176 - 177 .
L'acétyl-3 (chloro-3' propyl)-10 phénothiazine est une huile épaisse jaune-orange obtenue par condensation du chloro-1 bromo-3 propane sur l'acétyl-3 phénothiazine dans l'ammoniac liquide, en présence d'amidure de sodium préparé nin situ" et est utilisée brute.
EXEMPLE 5.
On opère comme dans l'exemple n 2 à partir de 10,3 g de trifluorométhyl-3 (chloro-3' propyl)-10 phénothiazine, 4,22 g de nipécotamide, 200 cm3 d'éthanol et 6,4 g au total de carbonate de sodium anhydre.
On chasse le solvant sous pression réduite (20 mm de mercure); on reprend le résidu par 50 cm3 d'eau et 150 cm3d'acétate d'éthyle; après agitation, on sépare la phase organique, on la sèche sur carbonate de potassium anhydre et on la concentre à sec sous pression réduite (20 mm de mercure).
Après lavage de la base cristallisée brute dans l'éther bouillant, on obtient 5,4 g de trifluorométhyl-3 (carbamoyl-3" pipéridino)-3' propyl]-10 phénothiazine, poudre cristalline blanche, fondant à 129-130 .
La trifluorométhyl-3 (chloro-3' propyl)-10 phénothiazine (P. F. : 74 - 76 ) est obtenue par condensation du chloro-1 bromo-3 propane sur la trifluorométhyl-3-phénothiazine dans l'ammoniac liqui- de, en présence d'amidure de sodium préparé "in situ".

EXEMPLE 6. -
On opère comme dans l'exemple n 2 à partir de 9,5 g de méthylthio-3 (chloro-3' propyl)-10 phénothiazine, 3,6 g de nipéco- tamide,200 cm3 d'éthanol et 5,3 g au total de carbonate de sodium anhydre.
La base brute huileuse, pesant 6,9 g, est dissoute dans 100 cm3 du mélange benzène-acétate d'éthyle (1: 1) et la solution est filtrée à travers une colonne de 100 g d'alumine pour chromato- graphie; on élue avec le même solvant. Après évaporation du solvant, on obtient 3 g de base purifiée que l'on recristallise dans l'acétate d'éthyle pour donner la méthylthio-3 [(carbamoyl-3" pipéridino)-3' propyl]-10 phénothiazine sous forme d'une poudre cristalline blanche fondant à 120-122 .
La méthylthio-3 (chloro-3' propyl)-10 phénothiazine est obtenue sous forme d'une huile épaisse jaune par condensation du chloro-1 bromo-3 propane sur la méthylthio-3 phénothiazine dans l'ammoniac liquide, en présence d'amidure de sodium préparé "in situ" et est utilisée brute.
EXEMPLE 7.-
On opère comme dans l'exemple n 2 à partir de 15,3 g de méthoxy-3 (chloro-3' propyl)-10 phénothiazine, 11 g de chlorhydrate de (N,N-diéthylearbamoyl)-4 pipéridine, 200 cm3 d'éthanol et 21,2 g au total de carbonate de sodium anhydre.
La base brute, huileuse, est purifiée par l'intermédiaire de l'oxalate acide que l'on prépare dans le méthanol; à partir de l'oxalate acide (poudre blanche fondant à 214-215 ), on revient à la base par mise en suspension dans l'eau, alcalinisation par un excès de soude et extraction par de l'acétate d'éthyle.
La base ainsi purifiée est transformée en chlorhydrate dans l'acétone par addition d'une solution éthérée d'acide chlorhy- drique ; on obtient ainsi 6,6 g de chlorhydrate de méthoxy-3 [(N,N-

On opère comme dans l'exemple n 2 à partir de 16 g de méthoxy-3 (chloro-3' méthyl-2' propyl)-10 phénothiazine, 11,8 g de chlorhydrate de morpholinocarbonyl-4 pipéridine, 200 cm3 d'éthanol et 21,2 g au total de carbonate de sodium anhydre.
La base brute est recristallisée successivement dans l'acéthonitrile et dans l'éthanol; on obtient ainsi 5,6 g de méthoxy- 3 (morpholino carbonyl-4" pipéridino)-3' méthyl-2' propyl]-10 phénothiazine, poudre cristalline blanche, fondant à 168-172 .
La méthoxy-3 (chloro-3' méthyl-2' propyl)-10 phénothia- zine est obtenue sous forme d'une huile épaisse jaune pâle en con- densant la méthoxy-3 phénothiazine sur le chloro-1 méthyl-2 bromo-3 propane dans l'ammoniac liquide, en présence d'amidure de sodium préparé "in situ" et est utilisée brute.
La morpholinocarbonyl-4 pipéridine (chlorhydrate, P. F. vers 305 ) est obtenue en hydrogénant dans le dioxanne en présence de Nickel Raney, à 170 sous 100 kg d'hydrogène, la morpholino- carbonyl-4 pyridine; ce dernier composé (P.F. : 75-76 ) est lui- même obtenu par action de la morpholine sur le chlorure d'isonico- tinoyle, chlorhydrate, dans le chloroforme à reflux.

The new phenothiazine derivatives can be converted into acid addition salts and quaternary ammonium derivatives.
The addition salts can be obtained by the action of the new derivatives on acids in suitable solvents. As organic solvents, for example alcohols, ethers, ketones or chlorinated solvents are used; water is advantageously used as inorganic solvent. The salt formed precipitates after possible concentration of its solution and is separated by filtration or decantation.
The quaternary ammonium derivatives can be obtained by the action of the new derivatives on esters, optionally in an organic solvent, at ordinary temperature or more rapidly by slight heating.
The novel phenothiazine derivatives of formula I can optionally be purified by physical methods, such as distillation, crystallization, chromatography, or chemical methods such as formation of salts, crystallization of the latter and decomposition of the latter in an alkaline medium. In these operations, the nature of the anion of the salt is irrelevant, the only condition being that the salt is well defined and easily crystallizable.
The new phenothiazine derivatives have interesting pharmacodynamic properties; in particular they are extremely active nervous system depressants, excellent

narcosis potentiators, tranquilizers, potent antiemetics and very good analgesics.
For therapeutic use, use is made of the new compounds either in the form of bases or in the form of addition salts or pharmaceutically acceptable quaternary ammonium derivatives, that is to say non-toxic. at the doses of use. The bases and addition salts defined above, however, constitute the preferred form of use, that is to say that in which the undesirable side actions are the weakest and the main action the strongest. .
As examples of pharmaceutically acceptable addition salts, there may be mentioned salts of mineral acids (such as hydrochlorides, sulphates, nitrates, phosphates) or organic acids (such as acetates, propionates, succinates, benzoates, fumarates, maleates, etc. oxalates, tartrates, methanesulfonates, ethanedisulfonates, chlorotheophyllinates, theophyllineacetates, salicylates, phenol-

As examples of pharmaceutically acceptable quaternary ammonium derivatives may be cited derivatives of mineral or organic acids such as chloro-, bromo- or iodomethylates, -ethylates, -allylates or -benzylates, methyl- or ethyl-. sulfates, benzenesulfonates or substitute derivatives of these compounds.
The new compounds can be administered neat or in the presence of a diluent or provided with a coating. The usual pharmaceutical forms can be used, in particular those which are suitable for oral, rectal or parenteral administration. The doses depend on the desired therapeutic effect, on the route of administration, on the duration of the treatment and on the animal species. They are generally between 0.1 and 10 mg per kilogram of animal weight. In human therapy, doses of 20 mg to 600 mg of active product by the oral route and 10 mg to 500 mg by the parenteral route can be accepted as daily doses.

Heated for 7 hours, at 2000, in an autoclave,

After cooling, the ammonia is evaporated off with slight warming and the mixture is then concentrated to dryness under a pressure of about 15 mm of mercury.
The residue is dissolved in 200 cm3 of ethyl acetate and the solution is exhausted with 100 cm3 of 0.7N methanesulfonic acid solution; the acidic aqueous phase is filtered through 1 g of charcoal and it is made alkaline with 20 cm3 of 4N sodium hydroxide solution; the base liberated is extracted with ethyl acetate, the solution is dried with anhydrous potassium carbonate and evaporated to dryness under a pressure of about 15 mm of mercury.
The solid residue thus obtained is finally recrystallized from acetonitrile; 12.5 g of methoxy-3 [(carbamoyl-4 "piperidino) -3 'propyl] -10 phenothiazine are thus obtained, a white-cream crystalline powder, melting at 132-134.

EXAMPLE 4.-
The operation is carried out as in Example 2 starting with 10 g of acetyl-3 (3-chloro-propyl) -10 phenothiazine, 4.45 g of isonipecotamide, 200 cm3 of ethanol and 7.4 g. in total of sodium carbonate - anhydrous.
After two successive recrystallizations of the crude base in ethanol, 4.55 g of acetyl-3 [(4-carbamoyl-piperidino) -3 'propyl] -10 phenothiazine, yellow-orange crystalline powder, mp: 176 are obtained. - 177.
Acetyl-3 (3-chloro-propyl) -10 phenothiazine is a thick yellow-orange oil obtained by condensation of 1-chloro-3-bromo propane with 3-acetyl phenothiazine in liquid ammonia, in the presence of sodium amide prepared in situ "and is used crude.
EXAMPLE 5.
The operation is carried out as in Example 2 starting with 10.3 g of trifluoromethyl-3 (3-chloropropyl) -10 phenothiazine, 4.22 g of nipecotamide, 200 cm3 of ethanol and 6.4 g in total. of anhydrous sodium carbonate.

EXAMPLE 6. -
The operation is carried out as in Example 2 starting with 9.5 g of methylthio-3 (3-chloro-propyl) -10 phenothiazine, 3.6 g of nipecotamide, 200 cm3 of ethanol and 5.3 g. in total of anhydrous sodium carbonate.
The crude oily base, weighing 6.9 g, is dissolved in 100 cm3 of the benzene-ethyl acetate mixture (1: 1) and the solution is filtered through a column of 100 g of alumina for chromatography; it is eluted with the same solvent. After evaporation of the solvent, 3 g of purified base are obtained which are recrystallized from ethyl acetate to give methylthio-3 [(3-carbamoyl-piperidino) -3 'propyl] -10 phenothiazine in the form of' a white crystalline powder, melting at 120-122.
The 3-methylthio (3-chloro-propyl) -10 phenothiazine is obtained in the form of a thick yellow oil by condensation of the 1-chloro-3-bromo propane on the 3-methylthio phenothiazine in liquid ammonia, in the presence of sodium amide prepared "in situ" and is used crude.
EXAMPLE 7.-
The operation is carried out as in Example 2 starting with 15.3 g of methoxy-3 (3-chloropropyl) -10 phenothiazine, 11 g of (N, N-diethylearbamoyl) -4 piperidine hydrochloride, 200 cm3 of ethanol and 21.2 g in total of anhydrous sodium carbonate.
The crude, oily base is purified by means of the acid oxalate which is prepared in methanol; starting from the acid oxalate (white powder melting at 214-215), the base is returned to the base by suspension in water, basification with excess sodium hydroxide and extraction with ethyl acetate.
The base thus purified is converted into the hydrochloride in acetone by adding an ethereal solution of hydrochloric acid; this gives 6.6 g of 3-methoxy hydrochloride [(N, N-

Methoxy-3 (3-chloro-2 'methyl-propyl) -10 phenothiazine is obtained in the form of a thick pale yellow oil by condensing 3-methoxyphenothiazine on 1-chloro-2-methyl-bromo. 3 propane in liquid ammonia, in the presence of sodium amide prepared "in situ" and is used crude.
Morpholinocarbonyl-4 piperidine (hydrochloride, P.
