PROCEDE POUR LA PREF ARATION DE 3-METHOXY-N-METHYL-MORPHINANES OPTIQUEMENT
ACTIVES 'ET DE SES SELS.
L'invention a trait à la préparation de 3-méthoxy-N-méthyl-mor- phinanes optiquement actives, à partir de 3-hydroxy-N-méthyl-morphinane ra- cémique.
Il a été établi que ce but peut être atteint par un procédé ca- ractérisé en ce que la D,L-3-hydroxy-N-méthyl-morphinane, dans un ordre de succession quelconque est, d'une part, séparée en les- antipodes optiques et, d'autre part, méthylée au groupe hydroxy-.
On. utilise- da préférence¯ 1-'un ou l'autre des deux modes opéra- toires, selon l'antipode désiré. Pour l'obtention de la (-)3-méthoxy-N- méthyl-morphinane ou de ses sels, il est avantageux. de- séparer d'abord la D'L-3-hydro-N-méthyl-morphinane à l'aide d'un acide optiquement actif, en particulier 1'cide D-tartrique, car dans l'eau le tartrate- de (-}-3- hydroxy-N-méthyl-morphinane cristalliser d'abord;
la base- laevogyre obtenue à partir de ce tartrate est méthylés au moyen d'un agent de- méthylation, de préférence 1'hydroxyde de phényl-triméthyl-ammonium et la (-)-3-méthyl- xy-N-métbyl-morphinane formée- est transformée-, le cas- échéant-, en les sels désirés
La (+)-3-méthoxy-N-méthyl-morphinane et ses sels sont préparés avantageusement par méthylation de la D,L-3-hydroxy-N-méthyl-morphinane au moyen d'un agent de- méthylatio, de préférence- l'hydroxyde- de- phényl-trimé thyl-ammonium, la D.L-3-méthylx-N-méthyl-morphinane formée est traitée avec un acide optiquement actif, de préférence l'acide D-tartrique pour produire le tartrate de (+)3-mthoxy-N-méthyl-morphinane qui cristallise- dans l'al- cool sous forme d'un se-1 difficilement soluble;
à partir du tartrate- on peut obtenir la base libre et le cas échéant, les sels de- cette dernière.
Les nouveaux composés peuvent être utilisés comme médicaments (contre la douleur et contre la toux).

On effectue la méthylation de 51,4 parties en poids de D,L-3-hy- droxy-N-métbyl-morphinane, selon le procédé décrit à l'exemple 1, au moyen d'une solution de méthylation préparée-à partir de 51,5 parties en poids de chlorure de phényl-trimétbyl-ammonium. La D.L-3-méthoxy-N-méthyl-morphina- ne est isolée sous forme de son bydrobromure. Point de-fusion avec 1 mol d'eau 92 - 94 C, sans eau 239 - 240 C.
La base, qui peut être séparée de la solution aqueuse au moyen de soude, fond à 81 - 83 C
On fait dissoudre 27,1 parties en poids de D,L-3-méthoxy-N-mé- tbyl-morphinane avec 15 parties en poids d'acide D-tartrique dans 150 par- ties en volume d'alcool chaud, on laisse refroidir et on ensemence avec du tartrate de (+)3-méthoxy-N-méthyl-morpinane La forme dextrogyre diffici- lement soulble dans l'alcool se sépare et est filtrée et lavée avec un peu d'alcool. (La forme laevogyre peut être isolée- de l'eau-mère, après avoir concentré cette dernière et éliminé autant de la forme dextrogyre que pos- sible, par cristallisation du résidu dans l'acétone}.
Le tartrate de (+)-3-méthoxy-N-Méthyyl-morphinane fond avec un mol d'eau à 195 - 196 C. [Ó]20 = + 30,6 (c = 1,5 dans l'eau). La base dextrogyre du point de fusion 108 - 109 C est obtenue du tartrate au moyen de soude. Elle peut être transformée en l'hydrobromure qui fond à 122 - 124 C.
[Ó]20 D = +27,6 (c = 1,5 dans l'eau).
REVENDICATIONS.
1) Procédé pour la préparation de 3-méthoxy-N-méthyl-morphinanes optiquement actives, caractérisé par le fait que la D,L-3-hydroxy-N-méthyl- morphinane est dans-un ordre-de succession quelconque, d'une part, séparée en les antipodes optiques et, d'autre part, méthylée au groupe hydroxy.
