Werkwijze en inrichting voor het oxigeneren van gist.
Deze uitvinding heeft betrekking op een werkwijze en inrichting voor het oxigeneren van gist, met andere woorden een werkwijze voor het toevoeren, respectievelijk toevoegen, van zuurstof aan de gist.
De uitvinding is vooral bedoeld voor het oxigeneren van gist bij de productie van bier, meer speciaal voor het oxigeneren van Saccharomyces cerevisiae, doch meer algemeen kan zij ook in andere toepassingen worden aangewend waarbij een gist dient te worden geoxigeneerd.
Het is bekend dat de gist die bij de productie van bier wordt aangewend na de hoofdgisting wordt afgescheiden van het bier, wat "het oogsten van de gist" wordt genoemd. De geoogste gist dient te worden behandeld, meer speciaal te worden geoxigeneerd, met andere woorden met zuurstof worden gevoed, om terug doeltreffend te kunnen worden aangewend in een volgende hoofdgisting.
De reden dat de gist dient geoxigeneerd te worden bestaat erin dat de gist zieh anders vanaf een gegeven ogenblik niet meer verder ontwikkelt. Wanneer gist tijdens een brouwproces in contact wordt gebracht met een voedingsstof, bijvoorbeeld wort, gaat de gist groeien. Deze gist produceert als nevenproduct ook Comet als gevolg dat na ongeveer l uur alle zuurstof uit het fermentatiemedium verdwenen is. De gist blijft evenwel groeien en deelt zich steeds verder op. De gist heeft evenwel slechts een bepaalde concentratie aan sterolen en onverzadigde vetzuren in zijn celmembraan, welke niet aangroeien, zodat per opdeling steeds minder sterolen en onverzadigde vetzuren

per nieuw gevormd celmembraan ter beschikking komen. Na een gegeven moment kan het celmebraan zich dan ook niet verder opdelen, waardoor de gist inactief wordt.
Dit kan hersteld worden door de gist te voeden met zuurstof.
Volgens een bekende methode wordt bij het aanvangen van een nieuwe hoofdgisting eerst de voedingsstof, bijvoorbeeld de wort, belucht en wordt dan de uitgeputte gist die afkomstig is van een vorige gisting in de beluchte voedingsstof gebracht, waardoor de ingevolge de beluchting in de voedingsstof aanwezige zuurstof dan voor de oxigenatie van de gist zorgt.
Een nadeel van deze bekende techniek bestaat erin dat verschillende ongewenste oxidatiereacties optreden die uiteindelijk in ongewenste kleur-en smaakveranderingen van het eindproduct resulteren. Bovendien is het op deze wijze onmogelijk om de juiste hoeveelheid toe te voegen zuurstof te bepalen omdat niet geweten is wat de huidige toestand is van de geoogste gist, met andere woorden niet geweten is in hoeverre de geoogste gist reeds uitgeput is, wat uiteindelijk het risico oplevert dat een overbeluchting of onderbeluchting wordt uitgevoerd, wat resulteert in een te grote groei of te geringe groei.
Deze bekende techniek heeft ook als nadeel dat de overdracht van zuurstof gering is ingevolge de schuimvorming die tijdens het beluchten ontstaat. Ook is deze techniek weinig doeltreffend in het geval van wort met een grote densiteit, omwille van het feit dat de zuurstof dan moeilijker in de wort oplost.
Volgens een andere bekende techniek wordt de geoogste gist verdund en door middel van een roerarm in beweging

gebracht, terwijl zuurstof via toevoerleidingen in de daartoe gebruikte tank wordt gebracht. Deze techniek heeft als nadeel dat, omwille van de verdunning en de schuimvorming, een grote tank noodzakelijk is. Bovendien moet ingevolge de verdunning nadien terug in een afscheiding van de gist worden voorzien.
Verder is het ook bekend om de gist te beluchten door lucht of zuurstof hierin te laten opborrelen. Deze techniek, waarbij gebruik wordt gemaakt van een zogenaamde "internal loop reactor", heeft als nadeel dat de overdracht van zuurstof bij gist met een hoge densiteit te gering is, en zelfs geringer is dan in het geval van een mechanische agitatie.
De oxigenatie van gist kan ook worden verwezenlijkt met behulp van een zogenaamde "external loop reactor", waarbij in een omloopcircuit door middel van een blaasmond een geforceerde zuurstofstroom in de gist wordt geblazen. Deze techniek heeft als nadeel dat een relatief grote druk moet worden gecreëerd, dat een grote schuimvorming optreedt, en dat de celwanden van de gist worden beschadigd door het impact van de zuurstof.
De uitvinding beoogt een werkwijze en inrichting voor het oxigeneren van gist die toelaten dat de voornoemde nadelen kunnen worden uitgesloten.
Hiertoe betreft de uitvinding in eerste instantie een werkwijze voor het oxigeneren van gist, waarbij zuurstof, hetzij zuivere zuurstof, hetzij zuurstof in gebonden toestand, aan een gistsuspensie wordt toegevoegd, met als kenmerk dat de toevoeging wordt verwezenlijkt door de

zuurstof via een poreuze membraanstructuur in de gistsuspensie te verdelen.
Hierdoor wordt het gas op een gelijkmatig verdeelde wijze en over een groot oppervlak in contact gebracht met de gistsuspensie, met als resultaat dat het gas gelijkmatig wordt opgenomen, met minder schuimvorming, en dat een zeer goede zuurstofoverdracht wordt verkregen.
Bij voorkeur wordt een starre poreuze membraanstructuur aangewend, die men een"membrane sparger"kan noemen, en wordt de gistsuspensie langs een bepaald gedeelte van het oppervlak hiervan bewogen, terwijl op een ander gedeelte van het oppervlak met een overdruk zuurstof op de membraanstructuur wordt geplaatst.
In de meest voorkeurdragende uitvoeringsvorm wordt de werkwijze verwezenlijkt door de gist te laten rondcirculeren via een omloopcircuit, dat bijvoorbeeld parallel is aangebracht over een tank waarin de gistsuspensie aanwezig is, en door de zuurstof aan de suspensie toe te voegen door middel van een poreuze membraanstructuur die zich in het omloopcircuit bevindt.
Door het rondcirculeren wordt een goede menging verkregen, waardoor een degelijke oxigenatie ontstaat van de volledige gistsuspensie.
In een voorkeurdragende uitvoeringsvorm wordt de doorstroomsnelheid van de gistsuspensie doorheen de membraanstructuur geoptimaliseerd door middel van een einmalige regeling en wordt de zuurstoftoevoer gedurende de oxigenatie geregeld. In het laatste geval geniet het de voorkeur dat de zuurstoftoevoer wordt geregeld in functie

van een meting waarmee de in de gist opgenomen zuurstof wordt gemeten.
De uitvinding heeft eveneens betrekking op een inrichting om de voornoemde werkwijze te verwezenlijken, waarvan de kenmerken uit de verdere beschrijving zullen blijken.
Met het inzicht de kenmerken van de uitvinding beter aan te tonen, is hierna als voorbeeld zonder enig beperkend karakter een voorkeurdragende uitvoeringsvorm beschreven, met verwijzing naar de bijgaande tekeningen, waarin : figuur 1 schematisch een inrichting volgens de uitvinding weergeeft ; figuur 2 op een grotere schaal een doorsnede weergeeft volgens lijn 11-11 in figuur 1 ; figuur 3 een doorsnede weergeeft volgens lijn 111-111 in figuur 2 ; figuur 4 een uitvergroting weergeeft van het gedeelte dat in figuur 2 met F4 is aangeduid ; figuur 5 een curve weergeeft die de zuurstofopname door de gist tijdens de oxigenatie voorstelt.
Zoals weergegeven in figuur'1 bestaat de inrichting 1 volgens de uitvinding hoofdzakelijk uit een tank 2 waarin de te oxigeneren gistsuspensie 3 wordt aangebracht en een omloopcircuit 4 dat in verbinding staat met de tank 2 en waarin een zuurstoftoevoerinrichting 5 is aangebracht met een poreuze membraanstructuur 6 via dewelke zuurstof aan de gistsuspensie 3 kan worden toegevoegd.
De membraanstructuur 6 bestaat zoals meer in detail is weergegeven in de figuren 2 tot 4 uit een lichaam 7 uit een poreus materiaal, waarbij in dit lichaam doorstroomkanalen

8 zijn aangebracht voor de doorstroming van de gistsuspensie 3, terwijl aan de buitenzijde 9 van het lichaam 7 een kamer 10 is gevormd waarin zuurstof onder druk wordt toegevoerd, via een toevoerleiding 11 die in verbinding staat met een zuurstofbron 12.
Voor het poreus materiaal wordt bij voorkeur gebruik gemaakt van aluminiumoxyde, daar dit inert is, een goede chemische weerstand biedt en mechanisch zeer sterk is.
Het voornoemde lichaam 7 bestaat zoals weergegeven bij voorkeur uit een stang waarin in langsrichting de doorstroomkanalen 8 zijn gevormd en vertoont in werkelijkheid bij voorkeur een lengte van 80 ä 100 cm, meer speciaal 90 cm en een diameter van 2 ä 3 cm, meer speciaal 2, 5 cm.
Hierin zijn bij voorkeur 15 ä 25 en meer speciaal 19 doorstroomkanalen 8 gevormd met een diameter van 2 ä 3 mm, en meer speciaal 2, 5 mm.
De poriën van de membraanstructuur 6 zijn bij voorkeur van de orde van grootte van 0, 02 tot 0, 2 micrometer.
Het voornoemde lichaam 7 is, bij voorkeur met de doorstroomkanalen 8 rechtop, in een behuizing 13 aangebracht.
Deze behuizing 13 vormt enerzijds een mantel 14 op een afstand rond het lichaam 7, zodat een kamer 10 zoals voornoemd ontstaat, en voorziet anderzijds in ruimten 15-16 die respectievelijk als een ingangscollector en uitgangscollector fungeren.
De ingang 17 van het omloopcircuit 4 bevindt zich onderaan aan de tank 2, terwijl de uitgang 18 bovenaan in de tank 2 uitgeeft.

In het leidinggedeelte 19 tussen de ingang 17 en de ruimte 15 is een pomp 20 aangebracht. Het pompdebiet van deze pomp 20 is bij voorkeur eenmalig instelbaar door middel van een regeleenheid 21.
Bij voorkeur zal de zuurstoftoevoer geregeld worden via een regelbaar ventiel 22. In het weergegeven voorbeeld gebeurt dit door middel van een regeleenheid 23 die voorziet in een regeling in functie van meetresultaten die bekomen worden door middel van een meetinrichting 24 die is aangebracht tussen de tank 2 en de zuurstoftoevoerinrichting 5, en die de zuurstofconcentratie in het medium meet.
De regeleenheid 23 is bij voorkeur zodanig ontworpen dat hiermee de opnamecapaciteit aan zuurstof van de gist kan worden opgevolgd. Deze opnamecapaciteit is weergegeven in de curve van figuur 5, waarbij in de ordinaat de opnamecapaciteit OUR is uitgezet in mg/min., terwijl in de abscis de tijd t is uitgezet in minuten.
Verder is deze regeleenheid 23 bij voorkeur ook zodanig ontworpen dat de zuurstoftoevoer wordt onderbroken nadat een welbepaalde tijd T is verstreken te rekenen vanaf het ogenblik dat zieh een top P in de curve heeft voorgedaan, welke tijd T bij voorkeur ongeveer twee uur bedraagt. Op dat ogenblik is immers de oxigenatie optimaal.
Verder is de inrichting 1 bij voorkeur zoals weergegeven voorzien van een koelinrchting 25, die er in het geval van bier voor zorgt dat de gistsuspensie 3 op een lage temperatuur wordt gehouden, bijvoorbeeld in de orde van grootte van 15 C.

De werking van de inrichting van figuur 1, alsmede de hiermee gepaard gaande werkwijze, is als volgt.
Vooreerst wordt de geoogste gist in de vorm van een gistsuspensie 3 in de tank 2 gebracht, bijvoorbeeld door middel van een toevoerleiding 26 waarin een pomp 27 is aangebracht.
Vervolgens wordt de voornoemde pomp 20 ingeschakeld, met een vooraf bepaald debiet, zodanig dat de gistsuspensie 3 via het omloopcircuit 4 in beweging wordt gebracht en alle gist gedwongen wordt door de zuurstoftoevoerinrichting 5 te stromen.
In de kamer 10 wordt zuurstof onder druk toegelaten. Zoals weergegeven in figuur 4 dringt deze zuurstof door de poriën van de membraanstructuur 6 en dringt zodoende door tot in de doorstroomkanalen 8, waar hij in contact komt met de gist. Omwille van het feit dat de zuurstof doorheen de poreuze structuur in contact gebracht wordt met de gist, wordt verkregen dat de zuurstoftoevoer zeer gelijkmatig geschiedt via het volledige binnenoppervlak 28 van de doorstroomkanalen 8, waardoor de in de inleiding genoemde nadelen zich niet kunnen voordoen.
Wanneer de regeleenheid 23 via de meetinrichting 24 waarneemt dat de voornoemde top P zich voordoet, zorgt deze regeleenheid 23 ervoor dat het ventiel 22 automatisch na het verstrijken van de voornoemde tijd T, die bij voorkeur twee uur bedraagt, wordt onderbroken. Hierna kan de gist terug worden aangewend voor een brouwproces of dergelijke.
Voor de oxigenatie zal bij voorkeur zuivere zuurstof worden aangewend, doch het is duidelijk dat ook zuurstof in

gebonden toestand of dergelijke kan worden aangewend, bijvoorbeeld in de vorm van lucht.
Alhoewel het aangewezen is om de oxigenatie uit te voeren door middel van een omloopcircuit 4, is het ook duidelijk dat dit niet noodzakelijk is. Zo bijvoorbeeld kan ook een poreuze membraanstructuur 6 in de tank 2 worden neergelaten of aangebracht via dewelke zuurstof in de tank 2 wordt gepompt. Deze poreuze structuur kan in dat geval zowel stil staan als bewegen, bijvoorbeeld langzaam ronddraaien.
Opgemerkt wordt dat het gebruik van het omloopcircuit 4 toelaat dat het geheel gemakkelijk modulair kan worden uitgebouwd door meerdere zuurstoftoevoerinrichtingen 5 in parallel te schakelen.
Volgens de uitvinding kan gewerkt worden met gistsuspensies 3 die tot 80 gram gist per liter bezitten. In de praktijk zal bij voorkeur gewerkt worden met suspensies die meer dan 60 gram gist per liter bevatten.
De huidige uitvinding is geenszins beperkt tot de als voorbeeld beschreven en in de figuren weergegeven uitvoeringsvorm, doch dergelijke werkwijze en inrichting voor het oxigeneren van gist kunnen in verschillende varianten worden verwezenlijkt zonder buiten het kader van de uitvinding te treden.

Method and device for the oxygenation of yeast.
This invention relates to a method and apparatus for the oxidation of yeast, in other words a method for supplying or adding oxygen to the yeast, respectively.
The invention is primarily intended for the oxygenation of yeast in the production of beer, more particularly for the oxygenation of Saccharomyces cerevisiae, but more generally it can also be used in other applications in which a yeast is to be oxygenated.
It is known that the yeast used in the production of beer is separated from the beer after the main fermentation, which is called "yeast harvesting". The harvested yeast must be treated, more specifically oxygenated, in other words fed with oxygen, in order to be used again effectively in a subsequent main fermentation.
The reason that the yeast has to be oxygenated is that otherwise the yeast no longer develops from a certain moment. When yeast is brought into contact with a nutrient, for example wort, during a brewing process, the yeast grows. This yeast also produces Comet as a by-product, as a result of which all oxygen disappears from the fermentation medium after about 1 hour. However, the yeast continues to grow and divides further and further. However, the yeast only has a certain concentration of sterols and unsaturated fatty acids in its cell membrane, which do not grow, so that per division less and less sterols and unsaturated fatty acids

A drawback of this known technique is that various undesired oxidation reactions occur which ultimately result in undesired color and taste changes of the final product. Moreover, it is impossible in this way to determine the correct amount of oxygen to be added because it is not known what the current state of the harvested yeast is, in other words it is not known to what extent the harvested yeast has already been exhausted, which ultimately leads to the risk. results in an over-aeration or under-aeration being performed, resulting in an overgrowth or undergrowth.

It is also known to aerate the yeast by bubbling air or oxygen into it. This technique, which makes use of a so-called "internal loop reactor", has the drawback that the transfer of oxygen in yeast with a high density is too low, and even less than in the case of a mechanical agitation.
The yeast oxygenation can also be effected by means of a so-called "external loop reactor", in which a forced oxygen flow is blown into the yeast in a bypass circuit by means of a blowing nozzle. The drawback of this technique is that a relatively high pressure has to be created, that a large foaming occurs, and that the cell walls of the yeast are damaged by the impact of the oxygen.
The invention contemplates a method and device for the oxidation of yeast which allow the above-mentioned drawbacks to be excluded.
To this end, the invention primarily relates to a method for the oxidation of yeast, in which oxygen, either pure oxygen or oxygen in the bound state, is added to a yeast suspension, characterized in that the addition is effected by

Good circulation is obtained by circulating, which results in a proper oxygenation of the entire yeast suspension.

The operation of the device of figure 1, as well as the associated method, is as follows.

According to the invention it is possible to work with yeast suspensions 3 which have up to 80 grams of yeast per liter. In practice, preference will be given to suspensions containing more than 60 grams of yeast per liter.
