Description
Medicinal diet composition for preventing and treating hyperuricemia and gout and preparation method thereof
TECHNICAL FIELD The invention belongs to the technical field of health care products, and particularly relates to medicinal diet composition for preventing and treating hyperuricemia and gout and preparation method thereof
BACKGROUND Hyperruriceemia (HUA) means that the blood uric acid level of men and women is higher than 420pmol/L and 360pmol/L on two different fasting days under normal purine diet. Long-term elevation of uric acid often leads to arthritis damage, gout and impaired renal function. The latest research shows that the prevalence rate of hyperuricemia in China is 16.85%, and the number of hyperuricemia patients has reached 236 million. At present, western medicines for lowering uric acid have great adverse reactions, often accompanied by obvious gastrointestinal reactions, liver and kidney function damage, neurotoxicity and muscle toxicity, and exfoliative dermatitis appears in severe cases. Traditional Chinese medicine treatment of this disease has the advantages of definite clinical efficacy, high safety and little toxic and side effects. Traditional Chinese medicine is considered to have a clear curative effect on hyperuricemia and gout. Besides clinical prescriptions, Chinese patent medicines are also recommended for the prevention and treatment of hyperuricemia and gout, such as Tongfengding Tablet, Tongfengshu Tablet, Compound Shenjin Capsule, Simiao Pill, etc. However, these are all pharmaceutical products, which need to be used under the guidance of clinicians or pharmacists. There are few related medicine and food homologous products. Therefore, the products with medicine and food to reduce uric acid and prevent gout have broad prospects.
SUMMARY The technical problem to be solved by the invention lies in overcoming the defects of the prior art and providing a medicinal diet composition for preventing and treating hyperuricemia and gout and a preparation method thereof. The invention provides a medicinal diet composition for preventing and treating hyperuricemia and gout, which is prepared from the following raw materials in parts by weight:1-15 parts of Cichorium intybus L., 1-15 parts of Symphytum officinale L, 1-20 parts of Phyllanthus emblica, 1-15 parts of Sophora japonica L., 1-20 parts of Pueraria lobata, 1-15 parts of Perilla frutescens , 1-10 parts of Chrysanthemum indicum L., 1-15 parts of Folium Mori, 1-15 parts of Prunus mume, 1-20 parts of tuckahoe, 1-20 parts of Lilium lancifolium , 0.1-1 part of folic acid, 0.1-1.5 parts of chitosan oligosaccharide, 0.5 parts of xylooligosaccharide. And the preparation method of the medicinal diet composition for preventing and treating hyperuricemia and gout comprises the following steps: Weighing raw materials according to the weight parts of the medicinal diet composition for preventing and treating hyperuricemia and gout, mixing Cichorium intybus L., Symphytum officinale L, Phyllanthus emblica, Sophora japonica L., Pueraria lobata, Perilla frutescens
, Chrysanthemum indicum L. flower, mulberry leaf, Prunus mume, poria cocos and Lilium lancifolium , soaking in 10-20 times of water for 15-60 minutes, heating and refluxing for 0.5-2 hours, and filtering with 80-200 meshes Obtaining a first extracting solution. Adding 10-20 times of water into the dregs, refluxing and extracting for 0.5-2 hours, and filtering with 80-200 meshes to obtain a second extracting solution. Mixing the first extracting solution and the second extracting solution, concentrating, and adding folic acid, chitosan oligosaccharide and xylooligosaccharide to obtain the medicated diet composition for preventing and treating hyperuricemia and gout. The medicated diet composition for preventing and treating hyperuricemia and gout of the present invention can eliminate turbidity and promote diuresis, reduce acid and prevent arthralgia. To prevent and treat hyperuricemia and gout, it has obvious inhibitory activity on adenosine deaminase (ADA) and xanthine oxidase (XOD), two key targets of uric acid production, and significantly enhances the expression of uric acid transporter, the core target of uric acid excretion. The medicinal diet composition for preventing and treating hyperuricemia and gout has the characteristics of clear curative effect, convenient use, clear action target and pharmacological effect, wide source of raw materials, low cost, simple preparation method, high patient compliance and the like, and is suitable for patients with hyperuricemia and ventilation.
BRIEF DESCRIPTION OF THE FIGURES Fig. 1 is an association network diagram of traditional Chinese medicine raw materials-active ingredients-action targets of a medicinal diet composition for preventing and treating hyperuricemia and gout provided by an embodiment of the present invention.
DESCRIPTION OF THE INVENTION In order to make the object, technical scheme and advantages of the present invention clearer, the present invention will be further described in detail with reference to the drawings and examples. It should be understood that the specific embodiments described herein are only used to explain the present invention, and are not used to limit the present invention. The embodiment of the invention provides a medicinal diet composition for preventing and treating hyperuricemia and gout, which is prepared from the following raw materials in parts by weight: 1-15 parts of Cichorium intybus L., 1-15 parts of Symphytum officinale L, 1-20 parts of Phyllanthus emblica, 1-15 parts of Sophora japonica L., 1-20 parts of Pueraria lobata, 1-15 parts of Perilla frutescens , 1-10 parts of Chrysanthemum indicum L., 1-15 parts of Folium Mori, 1-15 parts of Prunus mume, 1-20 parts of tuckahoe, 1-20 parts of Lilium lancifolium , 0.1-1 part of folic acid, 0.1-1.5 parts of chitosan oligosaccharide, 0.5-5 parts of xylooligosaccharide. Wherein, quercetin from Cichorium intybus L., Symphytum officinale L and Sophora japonica L. can inhibit the release of inflammatory factors such as TNFa and IL-1P, inhibit the activation of inflammatory corpuscles of NF-KB and NLRP3, and resist gout arthritis induced by monosodium urate crystals in rodents. Puerarin and kaempferol extract from Pueraria lobata can improve antioxidant function, inhibit xanthine oxidase, reduce uric acid formation and promote uric acid metabolism. Luteolin-7-o-glucoside, apigenin-7-o glucoside, luteolin and apigenin in Chrysanthemum indicum were also proved to be inhibitors of xanthine oxidase, with IC50 of 23.61, 38.80, 1.54 and 1.96pM respectively, suggesting that Chrysanthemum indicum indicum can inhibit the formation of uric acid and prevent hyperuricemia. Molecular docking experiments further confirmed that kaempferol, an active ingredient contained in many traditional Chinese medicines (such as Chennai, Mume and Sophora japonica L.), occupied the catalytic active site of xanthine oxidase and competitively and reversibly inhibited xanthine oxidase. The research of network pharmacology suggests that the medicinal diet composition for preventing and treating hyperuricemia and gout can intervene in various links of gout and hyperuricemia with multi-components, multi-targets, multi-channels and multi angles, and cooperatively prevent and treat this chronic disease. The prescription adopts all food-grade raw materials, which is safe, effective, low in production cost and reliable in prevention effect. It can effectively prevent and treat related symptoms caused by high uric acid gout, and is an accurate medicated diet product with clear main components and clear target points.
