In der USA-Patentschrift Nr. 3, 499, 078 ist eine Gruppe Antibiotika geoffenbart, die als Everninomicinantibiotika bezeichnet werden und die Everninomicine B, C und D umfassen, und ihre Herstellung durch Züchtung eines Mikroorganismus der species Mikromonospora carbonacea unter submersen aeroben Bedingungen.
Es wurde nunmehr gefunden, dass durch Reduktion dieser Everninomicine B, C und Danihrer Nitrogruppe und gewünschtenfalls weitere Umwandlung an dieser Stelle eine neue Gruppe Everninomicinderivate erhalten wird, wobei diese neuen Verbindungen wertvolle antibakterielle Wirksamkeit aufweisen.
Das erfindungsgemässe Verfahren ist demgemäss vor allem dadurch gekennzeichnet, dass ein Everninomi- ein B, C oder D-Ausgangsmaterial an seiner Nitrogruppe mit einem metallischen Reduktionsmittel, insbesondere Aluminiumamalgam in Gegenwart eines wässerigen niederen Alkanols, reduziert und gewünschtenfalls ein so gebildetes Hydroxylaminoderivat in das entsprechende Nitrose-oder Nitronderivat umgewandelt wird, und die erhaltenen Hydroxylamino-, Nitroso- oder Nitronderivate als solche oder in Form pharmazeutisch annehmbarer Salze isoliert werden.
Die erfindungsgemäss herstellbare neue Gruppe von Everninomicinderivaten der planaren Formel (I) wird somit durch die Nitrosoeverninomicine B, C und D, die Hydroxylaminoeverninomicine B, C, D, die Nitrone dieser Hydroxylaminoeverninomicine B, C und D und die pharmazeutisch annehmbaren Salze dieser Verbindungen gebildet.

Die Verbindungen der Formel (t) enthalten eine saure phenolische Hydroxylfunktion, die leicht in ein korrespondierendes pharmazeutisch annehmbares Salz umwandelbar ist. Typische pharmazeutisch annehm- bare Salze, die im Rahmen dieser Erfindung liegen, sind Metallsalze wie Alkalimetallsalze (beispielswei- se Natrium-und Kaliumsalze), Erdalkalisalze (beispielsweise Calciumsalze) und Aluminiumsalze, sowie Aminsalze wie dieTriniederalkylamin-, (z. B. Triäthylamin-), Procain-, Dibenzylamin-, N-Benzylbetaphen- äthylamino-, N, N'-Dibenzyläthylen-diamin-, N,N'-Bis-dehydroabietyl-äthylen-diamin-, N-Niederalkylpiperidin- (z. B. N-Äthylpiperidin-) und N-Methylglucaminsalze.
Bei der bevorzugten Ausführungsform des erfindungsgemässen Verfahrens wird ein Everninomicin B, C oder D als Ausgangsmaterial mit Aluminiumamalgam in einer Reaktionsmischung reduziert, die ein wässe- riges niederes Alkanol enthält, und das entsprechende Reduktionsprodukt isoliert. Das niedere Alkanol kann günstig eines mit bis zu 4 Kohlenstoffatomen sein und ist vorzugsweise Äthanol. Das genaue Verhältnis Alkanol zu Wasser ist nicht von ausschlaggebender Bedeutung, hängt allerdings von solchen Faktoren wie der Wahl des Alkanols und der Löslichkeit des entsprechenden Everninomicinausgangsmaterials im gewählten Alkanol/Wassersystem ab, und dieses Verhältnis kann in jedem gegebenen Fall einfach bestimmt werden.
Für das Äthanol/Wassersystem wurde herausgefunden, dass ein Vol,-Verhältnis 60: 40 bis 40 : 60 vorteilhaft anwendbar ist, und im besonderen wurde ein 50 : 50-Verhältnis verwendet. Günstig ist das Gew.-Verhältnis von Aluminiumamalgam (berechnet als Aluminium) zu dem Everninomicinausgangsmaterial etwa 0, 2 : 1.
Das Reduktionsverfahren kann günstig beiRaumtemperatur durchgeführt werden. Während der Reduktion des Everninomicinausgangsmaterials mit dem Aluminiumamalgam wird die Reaktionsmischung vorzugsweise in Bewegung gehalten, beispielsweise durch Rühren, und die Reaktion fortschreiten gelassen, bis das Amalgam verschwindet. Während die Reaktion in Gegenwart von Luft durchführbar ist, wird es doch für günstig gehalten, die Reduktion in einer inerten Atmosphäre von beispielsweise Stickstoff oder Argon durchzuführen.
Die Herstellung des Aluminiumamalgams erfolgt auf gewohnte Weise, im typischen Fall durch Ätzen von Aluminiumfolie mit Natriumhydroxyd und darauffolgendes ein-oder mehrmaliges Behandeln der so präparierten Folie mit einer Quecksilber-II-Chloridlöslll1g und endlich durch Waschen des so erhaltenenAluminiumamalgamproduktes. Die verwendete Aluminiumfolie ist vorzugsweise eine, die 98 bis 99, 5% Aluminium enthält, sowie Spuren mindestens eines der Metalle Kupfer, Silizium und Eisen ; vorteilhaft wurde eine Alu- miniumfolie angewendet, die im Handel unter den Namen Reynolds Wrap erhältlich ist, die im typischen Fall 99, 35% Aluminium und etwa 0, 12% Kupfer enthält.
Bei der Herstellung des Amalgams wird die Aluminiumfolie günstig in Quadrate mit 0, 5 cm Seitenlänge geschnitten und zu Kügelchen gerollt. Vorzugsweise sollte das Aluminiumamalgam vor der Verwendung frisch bereitet werden, um so das Reduktionsverfahren zu erleichtern.

Zu diesem Zweck wird eine Auftrennung einer Mischung der Everninomi- eine B, C oder D im typischen Fall durch chromatographische Technik durchgeführt, z.
B. durch Verwendung einer Silikagelsäule mit einem Benzol/Acetonsystemals EluierungsmitteL Die Reinigung kann unter Verwendung von Lösungsmittelfälltechniken und chromatographischen Techniken durchgeführt werden. So kann beispielsweise dieReinigung von Everninomiein D vorteilhaft durch Auflösen des Materials inAceton, Fällen des Materials durch Zusatz einer Petroläther/Diäthyläthermischung, Abfiltern des Niederschlages und mindestens einmaligem Wiederholen dieser Arbeitsschritte durchgeführt werden, wobei dieser Vorgehensweise eine chromatographische Reinigung unter Verwendung einer Silikagelsäule mit einem Benzol/Aceton/System als Eluierungsmittel geschlossen wird. Spezifische Vorgangsweisen werden anschliessend im Zusammenhang mit den Beispielen beschrieben.
Die als Ausgangsmaterial verwendeten gereinigten Everninomicine B, C und D werden vorzugsweise bei relativ niedrigen Temperaturen gelagert, beispielsweise bei +5 bis +100C, sowie unter Ausschluss von Wasser und in einer inerten Atmosphäre.
Wie sich aus der obigen Beschreibung ergibt, entspricht die Strukturformel der als Ausgangsprodukte verwendeten Everninomicine B, C und D der Formel (I) mit der Ausnahme, dass X in jedem Falle eine Nitrogruppe ist.
Das unmittelbare Verfahrensprodukt gemäss der Erfindung ist gewöhnlich eine Mischung, die das korrespondierende Nitroso- und/oder Hydroxylaminoeverninomicin enthält, wobei die Hydroxylaminoeverninomicinkomponente dominiert. Beispielsweise erhält man bei der Reduktion von Everninomicin D durch das spezifische Verfahren nach Beispiel 1 eine Gew.-%-Ausbeute, bezogen auf das Gewicht des Ausgangsmaterials, von 33% gereinigtem Hydroxylaminoeverninomicin D, sowie etwas weniger als 1 % gereinigtes Nitrosoeverninomicin D.
Die Isolierung und Reinigung des erhaltenen Nitrosoeverninomicins und Hydroxylaminoeverninomicins kann unter Verwendung bekannter Vorgehensweisen durchgeführt werden, wie Extraktion, Chromatographie, Umkristallisierung und Umfällung. Vorzugsweise wird die Abtrennung des Hydroxylaminoeverninomicins und der korrespondierenden Nitrosoverbindung aus der Mischung dieser Materialien durch chromatographische Techniken durchgeführt, wie Dünnschichtchromatographie, wobei im typischen Fall Silikagelplatten zur Anwendung kommen, mit irgendeinem geeigneten Entwicklungs lösungs m1ttel, wofür repräsentative Beispiele die Systeme Aceton/Benzol und Aceton/Äthylacetat/Benzol sind. Wird nach der Umkristallisierungstechnik vorgegangen, kann jedes geeignete Lösungsmittel verwendet werden.
Bei der Reinigung des Everninomicins C wurde Äthylacetat als Rekristallisationsmedium verwendet.
Die Nitronderivate können durch Reaktion der korrespondierenden Hydroxylaminoverbindung mitdem geeigneten Aldehyd in an sich bekannter Weise hergestellt werden, beispielsweise durch einfaches Mischen der Reaktionsteilnehmer in einem inerten, gerührten Lösungsmittel (z. B. Äthanol). Die Reinigung kannunter Anwendung bekannter Vorgehensweisen, vorzugsweise durch Chromatographie erfolgen.
Die gemäss der Erfindung erhaltenen pharmazeutisch annehmbaren Salze können unter Anwendungbekann- ter Vorgangsweisen hergestellt werden, so z. B. durch Reaktion von äquimolaren Mengen des geeigneten

triumhydroxyd) in wässeriger Lösung und durch Lyophilisieren der erhaltenen Mischung, wobei das gewünschte Salz erhalten wird. Die Verbindungen werden vorzugsweise unter wasserfreien Bedingungen in einer inerten Atmosphäre, wie z. B. unter Stickstoff oder Argon aufbewahrt, sowie bei relativ niedrigen Temperaturen, wie z. B. bei +5 bis +10 C. Auf diese Weise wird nicht wünschenswerte Oxydation (beispielsweise der Hydroxylaminofunktion) und Hydrolyse (der Orthoestergruppierungen) verhindert oder auf ein Minimum beschränkt.
Die Nitrosoeverninomieine B, C und D können auch durch Oxydation des korrespondierenden Hydroxyl- aminoeverninomicins hergestellt werden ; somit wird gemäss einem weiteren Aspekt der Erfindung das korrespondierende Hydroxylaminoeverninomicin mit einem geeigneten Oxydationsmittel oxydiert, wobei das gewünschte Nitrosoeverninomicinprodukt erhalten wird. Günstig wird das Ausgangsmaterial mit einem Alkalimetallhypobromit in einem aprotischen Lösungsmittel oxydiert.
Beispiele für verwendbare Alkalimetallhypobromite sind Kaliumhypobromit, Lithiumhypobromit und vorzugsweise Natriumhypobromit, während repräsentative Beispiele für aprotischeLösungsmittelDioxan, Di- äthylenglykoldimethyläther (auch Diglym genannt), Methanol, Äthanol und vorzugsweise Tetrahydrofuran sind.
Bei der Durchführung des Oxydationsverfahrens wird im typischen Fall eine lomige Gew.-/Vol.-Lösung des Ausgangsmaterials im aprotischen Lösungsmittel mit einem äquivalenten Volumen einer wässerigen Lösung von Natriumhypobromit behandelt (hergestellt wieinOrganicSynthesisCOLL., Bd. 4 [1963], S. 45). Wenn die Reaktion vollständig abgelaufen ist, was durch Dünnschichtchromatographie nachgewiesen wird, kann das erhaltene Nitrosoeverninomicinprodukt durch Zusatz von Wasser zur Reaktionsmischung, gefolgt durch

Extraktion mit einem geeigneten halogenierten Lösungsmittel, wie beispielsweise Methylenchlorid, isoliert werden. Eine weitere Reinigung kann dann durch Dünnschichtchromatographie durchgeführt werden. Im typi- schen Fall kann eine Gesamtausbeute von 60% erreicht werden.
Als Alternative zum Oxydationsverfahren mit Hypobromit kann das Hydroxylaminoeverninomicinaus- gangsmaterial der Oxydation durch Luft in einem alkalischen Lösungsmittel unterworfen werden, beispielsweise wird Luft durch eine Lösung des Ausgangsmaterials in verdUnntem wässerigem Natriumhydroxyd durch- perlen gelassen.
Wird die Umwandlung eines Nitrosoeverninomicinproduktes In das korrespondierende Hydroxylamin- everninomicinprodukt gewünscht, kann dies nach Vorgehensweisen durchgeführt werden, die für die Umwandlung einer Nitrosogruppe in eine Hydroxylaminogruppe bekannt sind.
Die gemäss der Erfindung erhaltenen neuen Verbindungen der Formel (1) zeigen ein enges Spektrum von in vitro antibakteriellerWirksamkeit gegen grampositive Bakterien (z. B. Staphylococcus aureus, Staph. 11631, Staph. W., Streptococcus pyogenes C, Strep. pyogenes C-203 und Bacillus subtilis) und sind weiterhin wirksam gegen gewisse gramnegative Bakterien (z. B. Neisseria gonorrhea und Neisseria meningitidis). Die Ver- bindungen der Formel (I) sind weiterhin brauchbar als Laboratoriumsreagentien zur Feststellung der Anwesenheit von enterischen gramnegativen Organismen sowie auch von Klebsiella pneumonia und Escherichia coli.
Im Hinblick auf ihre Wirksamkeit gegen grampositive und gramnegative Organismen können die Verbindungen der Formel (1) auch für Sterilisierungsausrüstungen verwendet werden, wie sie beispielsweise in Operationssälen und im Spitalsbereich angewendet werden.
Die bevorzugten Verbindungen sind solche, die von Everninomicin D abgeleitet sind, insbesondere Hydroxylaminoeverninomicin D und die pharmazeutisch annehmbaren Salze dieser Verbindung, insbesondere das Natriumsalz.
In diesem Zusammenhang soll darauf hingewiesen werden, dass die neuen Everninomicin D-Derivate den zusätzlichen Vorteil aufweisen, dass das Everninomicin D-Ausgangsmaterial beim Verfahren gemäss derUSAPatentschrift Nr. 3, 499, 078 das vorherrschende Produkt ist. Auf Grund ihrer Löslichkeit und ihrer Stab1l1- tätsfaktoren sind die pharmazeutisch annehmbaren Salze üblicherweise eine bevorzugte Form der Verbindungen gemäss der Erfindung. Repräsentative Salze sind das Natrium- und das N - Methylglucaminsalz der folgenden Verbindungen :

Ein Vergleich von Hydroxylaminoeverninomicin D, einer repräsentativen Verbindung gemäss der Erfindung, mit seiner Vorstufe, dem Everninomicin D ergibt, dass das Hydroxylaminoeverninomicin D eine dem Everninomicin D vergleichbare antibakterielle Wirksamkeit gegen grampositive Bakterien zeigt, aber schneller absorbiert wird, höhere Serumkonzentrationen ergibt, und auch schneller ausgeschieden wird als das Everninomicin D. Die verwendeten Versuchsbedingungen und-anordnungen und die erhaltenen Resultate werden im folgenden wiedergegeben.
Die Bestimmung der in vitro antibakteriellen Wirksamkeit gegen grampositive Mikroorganismen wurde durch Anwendung üblicher Röhrchenverdünnungsverfahren durchgeführt. In jedem Falle wurden 10 5-Ver- dünnungen von 24 h-Brühekulturen als Inoculum verwendet, wobei der Endpunkt nach einer Inkubationszeit von 24 h bei +370C in einem Difoo-Penassay-Brühe Medium (DifcoLabs., Detroit, Michigan) gewähltwurde.
Zur Bestimmung der in vitro Wirksamkeit gegen gramnegative Mikroorganismen wurde ein Plattenverdün- nungsverfahren angewendet mit einem Thayer-Martin-Medium und unter Anwendung des bekannten ThayerMartin-Verfahrens. Die in vivo antibakterielle Wirksamkeit bei Mäusen wird in der folgenden Tabelle 11 als Dosis wiedergegeben, die benötigt wird, um 50% der Testtiere zu schützen (PD ). Dazu wurden jeweils Gruppen von 7Mäu- sen (männliche Albinomäuse CF-1, jeweils mit einem Gewicht von 18 bis 20 g) verwendet, wobei jede Gruppe mit fünf verschiedenen Dosen behandelt und 10 Mäuse jeweils als Kontrolltiere verwendet wurden.
Jede Maus erhielt eine einzige subeutane oder orale Dose 1 h nach der intraperitonalen Infektion mit etwa 107 Organismen. Die Kontrollmäuse waren im allgemeinen nach 18 bis 24 h nach der Infektion tot. 48 h nach der Infektion wurden die Überlebenden der behandelten Gruppen bestimmt. Die DD -Werte in mg/kg wurden nach üblichen Verfahren ermittelt.

Die für die verschiedenen Verbindungen erzielbaren Serumhöchstkonzentrationen (Serumspitzenwerte) wurden durch Verabreichung einer einzigen Dose der Testverbindung an das Testtier bestimmt, wobei diese Dose 25 mg/kg für Ratten und 10 mg/kg für Hunde betrug. Nach der Verabreichung wurden inAbständen Blutproben gezogen und die Menge der Testverbindung im Serum in bekannter Weise bestimmt.

US Pat. No. 3,499,078 discloses a group of antibiotics called everninomicin antibiotics, including everninomicins B, C and D, and their preparation by culturing a microorganism of the species Mikromonospora carbonacea under submerged aerobic conditions.

The preferred compounds are those derived from everninomicin D, especially hydroxylaminoeverninomicin D and the pharmaceutically acceptable salts of this compound, especially the sodium salt.

As described in Examples 3 and 5, hydroxylaminoeverninomicin B or hydroxylaminoeverninomicin C is treated in anhydrous ethanol with benzaldehyde, 2-furfuraldehyde, and n-heptanal.
The products obtained are each isolated and purified in the manner described in Examples 3 to 5, whereby the benzyl nitrone, furfuryl nitrone and heptyl nitrone derivatives of hydroxylaminoeverninomicin B and hydroxylaminoeverninomicin C are obtained.
