Verfahren zur Darstellung von Monohalogenverbindungen gesättigter Ketone bzw. von unge- sättigten Ketonen der Pregnanreihe.
Die vorliegende Erfindung betrifft ein Verfahren zur Darstellung von Monohalogenverbindungen gesättigter Ketone bzw. von ungesättigten'Ketonen der Pregnanreihe, das darin besteht, dass man auf Pregnandione- (3. 20) bzw. auf Pregnanol- (20)-on- (3) oder dessen Derivate, die in 20-Stellung eine durch Hydrolyse in die Hydroxylgruppe umwandelbar Gruppe enthalten, insbesondere dessen Ester, die zur Bildung der Monohalogenverbindungen erforderliche Menge Halogen, zweckmässig Chlor oder Brom, vorzugsweise in Gegenwart eines organischen Lösungsmittels einwirken lässt und aus den entstehenden Monohalogenverbindungen gegebenenfalls Halogenwasserstoff in an sich üblicher Weise abspaltet, wobei man im Falle der Verwendung von Pregnanol- (20) -on- (3)
-derivaten der oben angegebenen Art als Ausgangsstoffe die in den entstehenden Verbindungen vorhandene, in die Hydroxylgruppe umwandelbare Gruppe in die Hydroxylgruppe-verwandelt.
Geht man bei dieser Umsetzung vom Pregnandion- (3. 20) aus, so gelangt man über die 4-Monohalogenverbindung zum Pregnen- (4)-dion- (3. 20), während man, ausgehend vom Allo-pregnandion- (3. 20), über die 2-Monohalogenverbindung zum Allo-pregnen- (1)-don- (3. 20) gelangt.
Die als Ausgangsstoffe für das vorliegende Verfahren in Betracht kommenden gesättigten ketogruppenhaltigen Verbindungen können in beliebiger Weise gewonnen sein, sei es durch direkte Isolierung aus natürlich vorkommenden Stoffen, sei es auf synthetischem Wege durch Umwandlung anderer Verbindungen.
Der Reaktionsverlauf sei beispielsweise durch die folgenden Formelbilder wiedergegeben, in denen CX eine Carbonylgruppe oder eine sekundäre Alkoholgruppe bzw. eine durch Hydrolyse in eine sekundäre Alkoholgruppe überführbare Gruppe bedeutet :

: Die Halogenierung und Halogenwasserstoffabspaltung erfolgt in an sich bekannter Weise in jeweils geeigneten Lösungsmitteln. Vorzugsweise erfolgt die Halogenwasserstoffabspaltung mittels Pyridinbasen.
Die nach dem vorliegenden Verfahren erhältlichen ungesättigten Ketone der Pregnanreihe stellen wertvolle Stoffe dar, die entweder selbst physiologisch wirksam sind oder sich auf physiologisch wirksame Stoffe weiterverarbeiten lassen.
Beispiel 1 : Zu einer Lösung von 1 g Pregnandion- (3. 20) in 50 cm3 Chloroform wird bei 0 eine Lösung von 0,5 g Brom in 20 cm3 Chloroform unter lebhaftem Rühren eingetragen. Das Chloroform wird im Vakuum abgedampft. Der Rückstand wird mit 10 cm3 Pyridin durch 12 Stunden unter Rückfluss gekocht. Hierauf wird die Reaktionsmischung in verdünnte Schwefelsäure gegossen und mit Äther extrahiert. Die ätherische Lösung wird mit Sodalösung und Wasser gewaschen, worauf der Äther abgedampft wird.
Der erhaltene Rückstand gibt bei weiterer Reinigung durch Umkristallisieren Pregnen- (4)-dion- (3. 20), das durch seine physiologische Wirkung sowie auch durch die Analyse und seine chemischen Eigenschaften sich als identisch erweist mit dem Corpusluteum-Hormon, wie es von Corpora-lutea der Eierstöcke isoliert wurde. Das ungesättigte Diketon kommt in zwei Modifikationen vor, deren eine bei 1210 und deren andere bei 128, 5 schmilzt : beide Modifikationen können leicht ineinander umgewandelt werden. Diese Umwandlung wird z. B. durch Auswahl entsprechender Lösungsmittel oder entsprechender Lösungsmittelgemische zum Umkristallisieren erreicht : so erhält man z.
B. die hochsehmelzende Modifikation, wenn man das Diketon aus wasserfreiem Methanol umkristallisiert. Dasselbe Produkt wird erreicht durch Wiederfestwerdenlassen des geschmolzenen Diketons, während das Umkristallisieren aus Alkoholwassermischungen immer Mischungen beider Modifikationen ergibt. Das niedrig schmelzende Produkt kann gewonnen werden durch Abdampfen einer Lösung des Diketons in Alkoholwasser-Äther-Mischung mittels eines Luftstromes.
Beispiel 2 : 2,05 g Pregnandion- (3. 20) werden in 30 cm3 Eisessig gelöst, hiezu werden 10 Tropfen einer Bromwasserstoff-Eisessig-Lösung und tropfenweise 3, 2cm3 einer Brom-Eisessig-Lösung [entsprechend 1,05 Mol. Brom] bei 15-200 unter Umrühren zugesetzt. Sofort beim Eintropfen der Bromlösung tritt Entfärbung ein. Das Reaktionsprodukt wird durch Zugabe von Wasser gefällt, abfiltriert und aus Alkohol sowie aus verdünntem Aceton umkristallidiert. Hiebei bildet es feine Nadeln mit seideartigem Glanz vom F =186-187 [unter Zersetzung]. Hierauf wird das reine oder rohe Produkt, wie es durch Fällung mit Wasser gewonnen wird, in ungefähr der 30-fachen Gewichtsmenge wasserfreien Pyridins unter Rückfluss durch etwa 15 Stunden gekocht.
Die Lösung wird in Wasser gegossen, das Pyridin wird durch verdünnte Schwefelsäure neutralisiert und mit Äther ausgezogen. Nach dem Abdampfen des Äthers verbleibt das Pregnen- (4)-dion- (3. 20), das nach Umkristallisieren oder einem andern Reinigungsprozess dem Corpus-luteum-Hormon entspricht. Es ist identisch mit dem nach Beispiel 1 erhaltenen ungesättigten Diketon : sein Dioxim schmilzt bei 244 [unter Zersetzung].
Beispiel 3 : 1, 5 g Pregnandion- (3. 20) werden in 100 cm3 Tetrachlorkohlenstoff gelöst und darauf unter Kühlung mit 50 cm3 einer 0,35 g Chlor enthaltenden Lösung von Chlor in Tetrachlorkohlenstoff versetzt. Nach längerem Stehen wird der Tetrachlorkohlenstoff im Vakuum verdampft und der erhaltene Rückstand mit ungefähr der 20-fachen Menge wasserfreien Chinolin 15 Stunden lang bei ungefähr 150-1600 erhitzt. Die weitere Aufarbeitung erfolgt wie in Beispiel 1 beschrieben, wobei ebenfalls Pregnen- (4)-dion- (3. 20) mit den Eigenschaften des Corpus-luteum-Hormons erhalten wird.
Beispiel 4 : 850 mg Allo-pregnandion- (3. 20) [F =200, 5 ] werden in 32 cm3 Eisessig gelöst.
Zu dieser Lösung gibt man 10 Tropfen einer Bromwasserstoff-Eisessig-Lösung und 1,27 cm3 einer Bromeislösung [entsprechend 1 Mol. Brom]. Die Reaktionslösung entfärbt sich sofort bei Zimmertemperatur.
Das 2-Brom-allo-pregnandion- (3. 20) wird durch Wasser gefällt und gibt durch Umkristallisieren aus Alkohol und verdünntem Aceton Nadeln vom F = 199 [unter Zersetzung].
150 mg dieses Monobromderivates werden durch 5 Stunden bei 17a-185 mit 5 cm3 einer 21 igen Lösung von Kaliumaeetat in Eisessig erhitzt. Das Reaktionsprodukt wird in Wasser gegossen und mit Äther extrahiert. Die ätherische Lösung wird mit Sodalösung und mit Wasser gewaschen, worauf der neutrale Anteil des erwähnten Ätherextraktes im Hochvakuum [0, 001 mm Hg] destilliert wird. Bei 800 sublimiert ein Teil der Substanz und gibt Kristalle, die nach dem Umkristallisieren aus

besitzen. Das Allo-pregnen- (l)-dion- (3. 20) zeigt im Ultraviolett eine Absorption bei etwa 235 mji, wie sie für a-ungesättigte Ketone charakteristisch ist.
Es ist nicht identisch mit dem Corpus-luteumHormon, aber isomer.
Während die Bromierung des Allo-pregnandions- (3. 20) ebenso einfach und glatt verläuft wie die des Pregnandions- (3. 20), unterscheidet sich das erhaltene 2-Brom-allo-pregnandion-(3. 20) von dem 4-Brom-pregnandion- (3. 20) wesentlich dadurch, dass sich aus ihm Bromwasserstoff nur schwer abspalten lässt. Man muss also, wenn man andere Halogenwasserstoff abspaltende Mittel verwenden will als das im obigen Beispiel angegebene, die Bedingungen für die Halogenwasserstoffabspaltung von Fall zu Fall vorher ermitteln.
Beispiel 5 : 500 mg Pregnanol- (20)-on- (3)-acetat werden in 4 cm3 Eisessig gelöst. Zu dieser Lösung werden 6 Tropfen einer Bromwasserstoff-Eisessig-Lösung und tropfenweise bei 20 0,7 cm3

einer Brom-Eisessig-Lösung entsprechend 1, 05 Mol. Brom} zugesetzt. Sofort tritt Entfärbung der Bromlösung ein. Wenn die Reaktion vollendet ist, wird das Reaktionsprodukt mittels Wasser gefällt und aus Alkohol umkristallisiert ; es bildet farblose Nadeln vom F = 167 .
310 mg des bromierten Acetats werden mit 5 cm3 wasserfreien Pyridins durch 14 Stunden zum Kochen erhitzt. Die Reaktionslösung wird dann durch Eindampfen konzentriert, mit verdünnter Schwefelsäure neutralisiert und mit Äther extrahiert. Nach dem Trocknen der ätherischen Lösung und dem Abdampfen des Äthers verbleibt ein öliger Rückstand, der durch Umkristallisieren aus wenig Alkohol gereinigt wird. Das kristalline Rohprodukt liefert beim Umkristallisieren aus verdünntem Alkohol das Acetat des ungesättigten Oxyketons in Form von Nadeln vom F = 138, 5 . Durch Verseifung mit l% iger methylalkoholisoher Alkalilauge erhält man daraus das Pregnen- (4)-ol- (20)-on- (3) vom F = 159 .
Beispiel 6 : Eine Lösung von 197 mg Chlor in 30 cm3 Chloroform wird unter Rühren und Kühlen einer Lösung von 1 g Pregnanol- (20)-on- (3)-aeetat in 50 cm3 Chloroform zugesetzt. Nach dem Abdampfen des Chloroforms wird der Rückstand mit 10 cm3 Chinolin durch 3 Stunden auf 150-1800 erhitzt. Die Reaktionslösung wird in verdünnte Schwefelsäure gegossen und die schwefelsaure Lösung mit Äther extrahiert. Der nach dem Abdestillieren des Äthers verbleibende Rückstand wird mit 25 cm3 einer l% igen alkoholischen Ätzkalilösung durch 2 Stunden erhitzt ; nach dem Abkühlen wird'in Wasser gegossen und mit Äther extrahiert.
Nach dem Verdampfen des Äthers kristallisiert man den Rückstand aus Alkohol um und erhält so das Pregnen- (4)-ol- (20)-on- (3) vom F = 1590.
Beispiel 7 : 7, 1 g Pregnanol- (20) -on- (3) werden in 100 em3 Eisessig gelöst, zu welcher Lösung tropfenweise eine Lösung der 1,02 Mol. entsprechenden Menge Brom in 12,5 cm3 Eisessig entspricht einer 4 n-Bromlösung] zugesetzt wird. Wenn man die Reaktionsmischung bei Zimmertemperatur stehen lässt, wird sie nach etwa 5 Minuten entfärbt. Das Reaktionsprodukt wird mit Wasser gefällt, abfiltriert und aus Alkohol und verdünntem Aceton umkristallisiert. Das so erhaltene Monobromderivat bildet feine Nadeln mit Seidenglanz vom F == 185 [unter Zersetzung].
550 mg dieses Monobromderivates werden mit 8 cm3 absolutem Pyridin 12 Stunden zum Sieden erhitzt. Die Aufarbeitung der Reaktionslösung geschieht, wie in Beispiel 5 angegeben. Das dabei erhaltene Pregnen- (4)-ol- (20)-on- (3) kristallisiert aus reinem Alkohol in charakteristischen Prismen,

Das nach diesem Beispiel erhältliche Monobromderivat ist identisch mit dem aus Pregnen- (4)- ol- (20)-on- (3) durch Bromwasserstoffanlagerung erhältlichen und kann ebenso wie dieses durch Oxydation und darauffolgende Bromwasserstoffabspaltung in Pregnen- (4)-dion- (3. 20) übergeführt werden (siehe österr. Patentschriften Nr. 154665, 155626 und 158271).
Die Abtrennung und Isolierung der monohalogenierten Zwischenprodukte und der ungesättigten Endprodukte lässt sich nicht nur in der Weise durchführen, wie in den Beispiel beschrieben, sondern man kann auch andere Methoden zur Anwendung bringen ; so kann man z. B. die Reaktionsgemische mit geeigneten Lösungsmitteln extrahieren und die letzteren abdampfen oder man kann die gebildeten Verbindungen aus ihren Lösungen mit Hilfe von Wasser oder organischen Lösungsmitteln, in denen sie unlöslich sind, ausfällen. Man kann sich ferner auch der Bildung unlöslicher oder schwerlöslicher Kondensationsprodukte, z. B. mit Ketonreagenzien u. dgl., bedienen.
Die Reinigung der genannten Zwischen-und Endprodukte kann auch durch fraktionierte und/oder wiederholte Kristallisation, Destillation, Sublimation ausgeführt werden.
PATENT-ANSPRÜCHE :
1. Verfahren zur Darstellung von Monohalogenverbindungen gesättigter Ketone bzw. von ungesättigten Ketonen der Pregnanreihe, dadurch gekennzeichnet, dass man auf Pregnandione- (3. 20), die zur Bildung der Monohalogenverbindungen erforderliche Menge Halogen, zweckmässig Chlor oder Brom, vorzugsweise in Gegenwart eines organischen Lösungsmittels einwirken lässt und aus den entstehenden Monohalogenverbindungen gegebenenfalls Halogenwasserstoff in an sich üblicher Weise abspaltet.

: The halogenation and elimination of hydrogen halide are carried out in a manner known per se in suitable solvents in each case. The hydrogen halide is preferably split off by means of pyridine bases.
The unsaturated ketones of the Pregnan range obtainable by the present process are valuable substances which are either physiologically active themselves or can be further processed into physiologically active substances.
Example 1: A solution of 0.5 g of bromine in 20 cm3 of chloroform is added at 0 to a solution of 1 g of pregnandione- (3. 20) in 50 cm3 of chloroform with vigorous stirring. The chloroform is evaporated in vacuo. The residue is refluxed with 10 cm3 of pyridine for 12 hours. The reaction mixture is then poured into dilute sulfuric acid and extracted with ether. The ethereal solution is washed with soda solution and water, whereupon the ether is evaporated.
On further purification by recrystallization, the residue obtained gives pregnen- (4) -dione- (3. 20), which proves to be identical to the corpus luteum hormone as it is from by its physiological effect as well as by analysis and its chemical properties Corpora lutea of the ovaries has been isolated. The unsaturated diketone occurs in two modifications, one melting at 1210 and the other melting at 128.5: both modifications can easily be converted into one another. This conversion is z. Discoloration occurs immediately when the bromine solution is added dropwise. The reaction product is precipitated by adding water, filtered off and recrystallized from alcohol and from dilute acetone. In doing so it forms fine needles with a silky sheen of F = 186-187 [with decomposition]. The pure or raw product, as obtained by precipitation with water, is then boiled under reflux in approximately 30 times the amount by weight of anhydrous pyridine for about 15 hours.
The solution is poured into water, the pyridine is neutralized with dilute sulfuric acid and extracted with ether. After the ether has evaporated, the pregnen- (4) -dione- (3. 20) remains, which corresponds to the corpus luteum hormone after recrystallization or another purification process. It is identical to the unsaturated diketone obtained in Example 1: its dioxime melts at 244 [with decomposition].
Example 3: 1.5 g of pregnandione (3. 20) are dissolved in 100 cm3 of carbon tetrachloride and then, while cooling, 50 cm3 of a solution of chlorine in carbon tetrachloride containing 0.35 g of chlorine are added. After standing for a long time, the carbon tetrachloride is evaporated in vacuo and the residue obtained is heated with about 20 times the amount of anhydrous quinoline for 15 hours at about 150-1600. The further work-up is carried out as described in Example 1, pregnen- (4) -dione- (3. 20) likewise being obtained with the properties of the corpus luteum hormone.
Example 4: 850 mg of allo-pregnandione- (3. 20) [F = 200, 5] are dissolved in 32 cm3 of glacial acetic acid.

150 mg of this monobromo derivative are heated for 5 hours at 17a-185 with 5 cm3 of a 21 solution of potassium acetate in glacial acetic acid. The reaction product is poured into water and extracted with ether. The ethereal solution is washed with soda solution and with water, whereupon the neutral portion of the ether extract mentioned is distilled in a high vacuum [0.001 mm Hg]. The reaction solution is then concentrated by evaporation, neutralized with dilute sulfuric acid and extracted with ether. After the ethereal solution has dried and the ether has evaporated, an oily residue remains, which is purified by recrystallization from a little alcohol. When recrystallized from dilute alcohol, the crystalline crude product yields the acetate of the unsaturated oxyketone in the form of needles with an F = 138.5. The pregnen- (4) -ol- (20) -one- (3) of F = 159 is obtained by saponification with 1% methyl alcoholic alkali.

The monobromo derivative obtainable according to this example is identical to that obtainable from pregnen- (4) -ol- (20) -one- (3) by addition of hydrogen bromide and, like this, can be converted into pregnen- (4) -dione- by oxidation and subsequent elimination of hydrogen bromide. (3. 20) are transferred (see Austrian patent specifications 154665, 155626 and 158271).
The separation and isolation of the monohalogenated intermediate products and the unsaturated end products can not only be carried out in the manner described in the example, but other methods can also be used; so you can z.
