Verfahren zur Darstellung von 5-Pyrazolonabkömmlingen.
Bekanntlich zeigen die l-Aryl-2-alkyl-pyrazolone- (5) bzw. ihre in 4-Stellung durch Aminogruppen substituierten Derivate fieberwidrige und schmerzstillende Eigenschaften.
Es wurde nun gefunden, dass, wenn man bei den für die Herstellung derartiger Pyrazolonderivate bzw. deren 4-Aminoabkömmlinge üblichen Verfahren von Hydrazinen ausgeht, welche an mindestens einem Stickstoffatom durch einen cycloaliphatischen bzw. hydroaromatischen Kohlenwasserstoffrest substituiert sind, zu Verbindungen nachstehender Formel gelangt :

Durch Methylierung mit Jodmethyl bei 1200 oder Dimethylsulfat bei 1700 erhält man daraus das 2. 3-Dimethyl-l-cyclohexylpyrazolon- (5), das nach dem Umlösen aus Essigester bei 660 schmilzt.
Es ist in Wasser und organischen Lösungsmitteln sehr leicht löslich.
33 Gewiehtsteile 2. 3-Dimethyl-l-cyclohexylpyrazolon- (5) werden in wenig Wasser gelöst, mit 100 Raumteilen 2n-Salzsäure versetzt und zu der auf 00 abgekühlten Lösung 100 Raumteile 2n-Natrium-. nitritlösung zugetropft. Es fällt die grün gefärbte 4-Nitrosoverbindung aus, die abgesaugt und mit den üblichen Reduktionsmitteln, wie Zinkstaub in salzsaurer Lösung oder katalytisch erregtem Wasserstoff, in die 4-Aminoverbindung übergeführt wird.
Das 4-Amino-2. 3-dimethyl-l-cyclohexylpyrazolon- (5) schmilzt nach dem Umlösen aus Essigester bei 1040 und ist in Wasser und Alkohol leicht löslich.

4-Dimethylamino-2. 3-dimethyl-l-cyclohexylpyrazolon- (5) ; es schmilzt nach dem Umlösen aus Benzin oder Hexahydrobenzol bei 77 und ist in Wasser sowie Alkohol leicht, in kaltem Benzin und Hexahydrobenzol schwer löslich.
Statt der Methylgruppen kann man in die 4-Aminogruppe auch einen oder zwei andere Reste, wie z. B. Benzyl-, Cyclohexyl-oder n-Hexylreste, ferner einen Benzyl-oder einen Cyclohexyl-und einen Methylrest einführen.
An das in 2-Stellung befindliche Stiekstoffatom kann statt der Methylgruppe auch ein anderer
Alkylrest, beispielsweise ein Äthyl-bzw. Isopropylrest oder ein cycloaliphatischer bzw. hydroaro- matischer Rest gebunden sein. Geht man von Homologen des Acetessigesters, beispielsweise vom
Methylacetessigester aus, so erhält man Pyrazolone, die in 4-Stellung durch einen Alkylrest sub- stituiert sind.
Setzt man 2 Mol 4-Amino-2. 3-dimethyl-l-cyclohexylpyrazolon- (5) in benzolischer Lösung mit l Mol 3-Bromcyclohexen- (l) um, so erhält man nach Abfiltrieren des ausgefallenen Hydrobromids des Ausgangsstoffes und Abdestillieren des Benzols das 2. 3-Dimethyl-l-cyclohexyl-4-[cyclohexen- (2')-ylamino]-pyrazolon- (5).
Die in 4-Stellung monomethylierte Verbindung erhält man aus dem 4-Amino-2.3-dimethyl- l-cyclohexylpyrazolon- (5) durch Anlagerung von Dimethylsulfat an die 4-Benzyliden-Verbindung und Abspaltung von Benzaldehyd (vgl. deutsche Patentschrift Nr. 267 700). Sie schmilzt nach dem Umlösen aus einem Gemisch von Hexahydrobenzol und Benzin bei 1020 und zeigt ähnliche Löslichkeitverhältnisse wie die 4-Dimethylaminoverbindung.
Beispiel 2 : 100 Gewiehtsteile NN'-Dieyclohexylhydrazin und 70 Gewichtsteile Acetessigester werden 6 Stunden lang auf 160-1800 erwärmt. Nach dem Abkühlen versetzt man mit Essigester und alkoholischer Salzsäure bis zur stark kongosauren Reaktion, worauf das Hydrochlorid des 3-Methyl- 1. 2-dicyclohexylpyrazolons- (5) ausfällt. Die freie Base schmilzt nach dem Umlösen aus Benzin bei 85 und ist in Wasser schwer, in Alkohol leicht löslich. Durch Nitrosierung und Reduktion analog Beispiel 1 erhält man aus ihr das 4-Amino-3-methyl-1. 2-dieyclohexylpyrazolon- (5), dessen Hydrochlorid nach dem Umlösen aus salzsäurehaltigem Alkohol bei 2050 schmilzt.
20 Gewichtsteile dieses Hydrochlorids werden mit Formaldehyd und Ameisensäure 3 Stunden auf dem Dampfbad erwärmt. Nach dem Abkühlen scheidet man die Base durch Zusatz von Alkalihydroxyl ab, nimmt in Äther auf und versetzt den Ätherrückstand mit Essigester und überschüssiger alkoholischer Salzsäure. Es kristallisiert das Hydrochlorid des 4-Dimethylamino-3-methyl-1. 2-dicyclo- hexylpyrazolon- (5) aus, das nach dem Umlösen aus salzsäurehaltigem Alkohol bei 2060 schmilzt. Die freie Base schmilzt nach dem Umlösen aus Petroläther bei 83 und ist in Wasser schwer, in Alkohol leicht löslich :
Auch hier lassen sich die in Beispiel 1 erwähnten Reste in 3-Stellung sowie in die 4-Aminogruppe einführen.
So kann man z. B. aus 4-Amino-3-methyl-1. 2-dicyclohexylpyrazolon- (5) das 4-Methyl- amino-3-methyl-1. 2-dieyclohexylpyrazolon- (5) erhalten, das bei der Umsetzung mit Formaldehydbisulfit eine wasserlösliche Formaldehydbisulfit-Verbindung liefert.
Beispiel 3 : Einfach molekulare Mengen Natriumacetat und Cyclopentylhydrazinhydrochlorid (F = 130 ) werden in der zehnfachen Menge Wasser bei 80 mit der berechneten äquimolekularen Menge Acetessigester 2 Stunden lang gut durchgerührt. Das entstandene 3-Methyl-l-cyclopentyl- pyrazolon- (5) wird abgesaugt ; es schmilzt nach dem Umlosen aus Alkohol bei 139 . Durch Methylieren mit Jodmethyl bei 120 oder Dimethylsulfat bei 1700 erhält man das 2. 3-Dimethyl-l-cyclopentylpyrazolon- (5) vom Kp3 = 161-163 .
36 Gewichtsteile des letzteren werden sodann in wenig Wasser gelöst, mit 120 Raumteilen 2n-Salzsäure versetzt und zu der auf 0'abgekühlten Lösung allmählich 100 Raumteile 2n-Natriumnitritlösung zugetropft. Die grün gefärbte 4-Nitrosoverbindung wird abgesaugt und mit den üblichen Reduktionsmitteln in das 4-Amino-2. 3-dimethyl-1-eyclopentylpyrazolon-(5) übergeführt (Kp4= 178-1820), welches nach dem Umlösen aus Essigester + Hexahydrobenzol bei 630 schmilzt. Mit Methylierungsmitteln, wie z. B. Formaldehyd und Ameisensäure, erhält man aus der 4-Aminoverbindung das 4-Dimethylamino-2'3-dimethyl-l-cyclopentylpyrazolon- (5) vom KPa = 1600.
Auch in diesem Beispiel können ebenso wie im Beispiel 1 Reste der dort angegebenen Art in die 4-Aminogruppe bzw. in 2-oder 3-Stellung eingeführt werden.
