Abstract

Reivindicacion 1: Un método para producir un anticuerpo monoclonal que comprende: a) proveer un hongo heterocarion que incluye un primer nucleo y un segundo nucleo, donde dicho primer nucleo incluye una primera construccion de ácido nucleico que incluye una primera secuencia de ácido nucleico que codifica para cadena liviana de un anticuerpo, y donde dicho segundo nucleo incluye una segunda construccion de ácido nucleico que incluye una primera secuencia de ácido nucleico que codifica para una cadena pesada de un anticuerpo, y donde por lo menos una de dichas construcciones de ácido nucleico comprende además una segunda secuencia de ácido nucleico que codifica para un polipéptido o para una parte funcional del mismo secretada normalmente por un hongo; b) cultivar el hongo heterocarion bajo condiciones apropiadas para la expresion de las cadenas livianas y pesadas del anticuerpo.It refers to methods for producing a monoclonal antibody in a heterocarion fungus or in a fungal host cell. Claim 1: A method of producing a monoclonal antibody comprising: a) providing a heterocarion fungus that includes a first nucleus and a second nucleus, wherein said first nucleus includes a first nucleic acid construct that includes a first nucleic acid sequence encoding for light chain of an antibody, and wherein said second nucleus includes a second nucleic acid construct that includes a first nucleic acid sequence encoding a heavy chain of an antibody, and wherein at least one of said nucleic acid constructs comprises furthermore a second nucleic acid sequence encoding a polypeptide or a functional part thereof normally secreted by a fungus; b) cultivate the heterocarion fungus under appropriate conditions for the expression of the light and heavy chains of the antibody.
