Definitions
- the present invention relates to a method and an apparatus for acquiring a tomographic image of a measurement target such as a biological tissue using the coherence of light waves.
- OCT Optical Coherence Tomography
- Patent Document 1 The measurement principle of acquiring a tomographic image using the coherence of light waves is called OCT (Optical Coherence Tomography), and is currently in practical use as a medical device for acquiring a tomographic image of the retina (for example, Patent Document 1). And Non-Patent Document 1).
- this device low coherence light is separated into measurement light and reference light, an object to be measured is installed on the measurement light side, a reference mirror is installed on the reference light side, and the reflected light from both is superimposed to produce an interference signal
- a tomogram is acquired by obtaining Therefore, the lower the coherence, that is, the wider the spectral width of the light source, the higher the depth resolution (hereinafter, resolution) can be obtained.
- this equipment can be broadly classified into time domain OCT (TD-OCT) and spectral domain OCT (SD-OCT: Spectral-domain OCT).
- TD-OCT time domain OCT
- SD-OCT spectral domain OCT
- OCT has a feature that the resolution decreases when the group velocity dispersion (hereinafter referred to as dispersion) existing in the measurement optical path and the reference optical path is different. Therefore, if there is dispersion in the object to be measured and the optical system in the vicinity thereof, there is a drawback that the resolution is lowered due to the influence.
- Non-Patent Document 2 a spectral intensity coherence tomographic imaging apparatus has been developed that automatically cancels the influence even when the dispersion existing in the measurement optical path and the reference optical path is different.
- the principle is similar to that of SD-OCT, but it has a slightly more complicated and large-scale device configuration compared to normal SD-OCT, such as requiring two spectrometers.
- the measurement by OCT has a feature that the resolution decreases when the dispersion existing in the measurement optical path and the reference optical path is different. For this reason, if there is dispersion in the object to be measured and the optical system in the vicinity thereof (that is, in the measurement optical path), the resolution is lowered.
- a method has been used in which the same dispersion as that existing in the measurement optical path is intentionally introduced into the reference optical path to cancel the effect (see, for example, Patent Document 2). .
- the present invention as with the SD-OCT device, as a method for realizing this with one spectroscopic device, a case where no phase difference is introduced between the measurement light and the reference light, and a case where a phase difference of ⁇ is introduced.
- the intensity of the two spectral interference fringes (the interference signal from the broadband light is wavelength-resolved by the spectroscopic device and becomes an interference fringe) is acquired in time series, and necessary calculations are performed based on the two intensities to obtain the tomographic information. Methods and apparatus for obtaining are provided.
- the present invention extends the above-described analysis method of spectral interference fringe intensity data acquired by two measurements with a phase difference, and is equivalent to analyzing spectral interference fringe intensity data by simpler one measurement.
- a method and apparatus for acquiring tomographic information having resolution is provided.
- the phase difference with respect to light having a wavelength other than the center wavelength does not strictly become ⁇ , and therefore an error occurs as the spectrum of the light source becomes wider.
- a method for generating a similar phase difference by controlling the polarization states of the measurement light and the reference light, a change in the geometric phase associated therewith can be used (for example, see Non-Patent Document 3). Since this method allows phase shift independent of the spectrum, an error depending on the spectrum width of the light source does not occur.
- the entire device can be realized at low cost.
- the optical adjustment becomes simpler than the conventional apparatus configuration using two spectroscopic apparatuses, and the factors causing measurement errors can be reduced.
- each arrow represents an optical path.
- the converted value of pixel pair length of the present invention (b) is half that of the SD-OCT device (a).
- FIG. (b) is a diagram showing the same experimental results when a dispersion medium (heavy flint glass) is installed in the measurement optical path immediately before the object to be measured.
- the converted value of the pixel pair length in (a) and (b) is half that of the SD-OCT device, FIG. (a) is a signal acquired before moving the detector, and (b) is a signal acquired after moving the detector a distance of about 6 pixels.
- the pixel-to-length conversion value is half that of the SD-OCT device, FIG.
- a mechanism or means for generating a phase difference ⁇ between measurement light and reference light is added to an existing SD-OCT apparatus (Example 1) or not added (Example 3), and the principle is realized.
- a tomographic image of the object to be measured is created by performing arithmetic processing (according to equations (2) to (4)).
- FIG. 1 shows an optical system of a spectral intensity coherence tomographic imaging apparatus proposed by the present invention.
- each arrow represents an optical path.
- This optical system is basically the same as a conventional SD-OCT device, but the reference mirror is placed on a fine movement stage driven by a piezo element so that the incident light strikes vertically, and the fine movement stage propagates the light. The difference is that it can move back and forth along the direction.
- the spectroscopic device is configured by the combination of the diffraction grating G and the detector Detector, but the same applies to other types of spectroscopic devices, generally combinations of spectroscopic elements and detectors.
- the fine movement stage is not limited to driving by a piezo element.
- the beam splitter may be a demultiplexer / multiplexer, and can be configured using a 2 ⁇ 2 optical fiber coupler.
- Light from a broadband light source having a wide spectral width is separated into reference light and measurement light by a beam splitter BS1.
- the light source is, for example, a superluminescent diode or a supercontinuum light source, but may be another type of light source as long as the spectrum is broadband.
- the reference light is reflected by the reference mirror, the measurement light is reflected by the object to be measured, and the respective return lights are combined and interfered by the beam splitter BS1.
- the interference light is decomposed into a spectrum by the diffraction grating, and the spectrum interference fringe intensity is detected by the detector.
- the reference mirror is installed on the fine movement stage.
- the spectral interference fringe intensity before moving the fine movement stage is detected and stored in a computer (with a display device).
- a computer with a display device.
- the intensity is recorded as a function of the wavelength ⁇ in the detector.
- the center frequency ⁇ 0 of the incident light is adjusted so as to coincide with the center of the detector.
- the fine movement stage is moved forward or backward along the light wave propagation direction by a distance that is half the center wavelength of the incident light (that is, the phase of the reference light is shifted by ⁇ ), and the same as above.
- the spectral interference fringe intensity is detected and stored in a computer (with a display device), and the same conversion is performed. This is I 2 ( ⁇ ).
- the spectral interference fringe intensity at the frequency shifted in the positive direction by the center frequency ⁇ 0 to ⁇ ′ is I 1 ( ⁇ 0 + ⁇ ′), and after the phase shift, the center frequency ⁇ 0 to ⁇ ′.
- the spectral interference fringe intensity at a frequency shifted in the negative direction is I 2 ( ⁇ 0 ⁇ ′).
- ⁇ ′ As a function of ⁇ ′, Fourier transform is performed on ⁇ ′ in a computer (with a display device). As a result, although the Fourier transform of C ( ⁇ ′) is generally a complex number, a target signal having information about the position of the object to be measured and reflectance is obtained in its real part. On the other hand, when the absolute value of the Fourier transform of C ( ⁇ ′) is evaluated, an unnecessary signal adjacent to the target signal is also confirmed, but this appears in the imaginary part, so that both can be easily separated. Is possible.
- FIG. Fig. 3 (a) shows the experimental results obtained with the conventional SD-OCT device when a simple reflecting mirror is installed as the object to be measured
- Fig. 3 (b) shows the experimental results obtained with the proposed device. ing.
- the reference mirror is placed on the piezo-drive fine movement stage and moved back and forth, but the object to be measured may be placed on the fine movement stage.
- ⁇ phase difference may be introduced (see Non-Patent Document 3). Furthermore, when performing the above calculation, how to take the spectral interference fringe intensity before and after the phase shift,
- FIG. 4 shows the same experimental results when a heavy flint glass, which is an example of a dispersion medium, is installed in the measurement optical path immediately before the object to be measured in order to evaluate the influence of dispersion of the proposed apparatus.
- FIG. 4A shows the result obtained by the conventional SD-OCT apparatus
- FIG. 4B shows the result obtained by the proposed apparatus.
- FIG. 4 (a) the signal of the conventional SD-OCT is spread due to the influence of dispersion and causes a reduction in resolution.
- FIG. 4 (b) of the present invention the width of the target signal is as shown in FIG. 3 (b). It can be seen that no change due to the dispersion is observed at all, and the resolution is not lowered.
- a reference mirror is installed on a fine movement stage driven by a piezo element so that the fine movement stage can be moved back and forth along the light propagation direction.
- this fine movement stage is not necessary, and it is sufficient that a general reference mirror distance adjustment mechanism (not shown, for example, a moving stage) provided in a conventional SD-OCT apparatus is provided.
- the reference mirror is fixed to the fine movement stage for convenience.
- the intensity of the spectral interference fringes detected by the detector adjusted so that the center frequency ⁇ 0 of the incident light coincides with the center of the detector is measured by the computer (with display device). ).
- this spectral interference fringe intensity is positive from the center frequency ⁇ 0 to ⁇ ′ with respect to the spectral interference fringe intensity I ( ⁇ ) converted to be a function of the frequency ⁇ using Equation 1.
- the spectral interference fringe intensity at the frequency shifted in the direction of is I ( ⁇ 0 + ⁇ ′), and the frequency shifted from the center frequency ⁇ 0 in the negative direction by ⁇ ′ (negative sideband)
- the intensity of the spectral interference fringe in the wave) is I ( ⁇ 0 ⁇ ′).
- ⁇ ′ As a function of ⁇ ′, Fourier transform is performed on ⁇ ′ in a computer (with a display device). As a result, the Fourier transform of C ( ⁇ ′) is always a real number, and unnecessary signals are also confirmed adjacent to each other.
- the position of the reference mirror is adjusted by adjusting the position of the reference mirror. A target signal having information on the position of the measurement target and reflectance can be obtained separately from unnecessary signals.
- the height of the signal is proportional to the square of the reflectance of the object to be measured (reflection mirror).
- the target signal and unnecessary signals can be separated. It is possible to acquire only a target signal having information on a measurement target.
- FIG. 6B shows the same experimental results when a heavy flint glass, which is an example of a dispersion medium, is installed in the measurement optical path immediately before the object to be measured.
- the corresponding experimental results obtained with a conventional SD-OCT apparatus are shown in FIG.
- the target signal width is the same as the result shown in FIG. 6A, and it can be seen that there is no change affected by the dispersion. Therefore, the resolution does not decrease.
- the target signal can be acquired separately from unnecessary adjacent signals by adjusting the position of the reference mirror as described above.
- Non-Patent Document 2 In the theory of the spectral intensity coherence tomographic imaging apparatus (see Non-Patent Document 2), it is clear that the size of the artifact varies depending on the center frequency ⁇ 0 of the incident light.
- the detector incorporated in the spectroscopic apparatus is arranged in the spectral dispersion direction and is about several percent of the detector size. Move within the range.
- FIG. 7 shows the experimental results obtained with the proposed apparatus when a microscope cover glass is used as the measurement target having the two discrete reflecting surfaces described above. In FIG. 7, only a target signal having information on the measurement target including artifacts is displayed.
- FIG. 7 (a) is a signal acquired before moving the detector
- FIG. 7 (b) shows a detector having 2048 pixel structures in the spectral dispersion direction, and about 6 pixels along the direction. It is a signal acquired after moving the distance.
- FIG. 7A it is confirmed that there is an artifact at the center in addition to the signals corresponding to the two reflecting surfaces to be measured.
- FIG. 7B it is confirmed that only the central artifact is reduced and the signals corresponding to the two reflecting surfaces to be measured hardly change.
- the data processing in the computer was performed by creating a program in the Visual C ++ (registered trademark) language.
- Visual C ++ registered trademark
- any type of information processing apparatus can be used. You may create and implement a program by language.

Abstract
Description
これをI1(ω)とする。
