Claims (1)
- 【特許請求の範囲】 1.カンピロバクター・コリ(Campylobacter coli)種に属する株のゲノム核 酸と特異的にハイブリダイズし、該株に属さないカンピロバクテリアの核酸とも 、カンピロバクター属に関連するバクテリアのゲノム核酸とも通常の条件下でハ イブリダイズしない核酸配列であって、該配列が、配列番号:1、それに相補的 な配列、又は1以上の塩基の変異、挿入、欠失もしくは置換によりそれらから異 なる配列からなる群から選択されるヌクレオチド配列を有することを特徴とする 核酸配列。 2.請求項1に記載のヌクレオチド配列を含むクローニングベクター。 3. No.I−1491下で1994年11月14日にナショナル・コレクション・オブ・ マイクロオルガニズム・カルチャーズに寄託されたプラスミド。 4.請求項1に記載のヌクレオチド配列の中から選択される少くとも20の連続 ヌクレオチドを含むカンピロバクター・コリの核酸の特異的核酸プローブ。 5.前記プローブが支持体上に固定化され、捕獲プローブとして用いられるこ とを特徴とする請求項4に記載の核酸プローブ。 6.カンピロバクター・コリの核酸配列を増幅するための特異的オリゴヌクレ オチドプライマーの対であって、該対が、請求項1に記載の配列の中から選択さ れる18〜30ヌクレオチド、好ましくは18〜22ヌクレオチドを有するオリゴヌクレ オチドの対を含むことを特徴とする対。 7.配列: のオリゴヌクレオチド対により形成されることを特徴とする請求項6に記載のカ ンピロバクター・コリ の核酸配列を増幅するための特異的オリゴヌクレオチドプ ライマーの対。 8.請求項6又は7に記載のオリゴヌクレオチドプライマーの対の手段により 、遺伝子増幅技術により、特にPCR技術によりC.コリの核酸を増幅することに より得られる核酸フラグメント。 9.生物試料中のカンピロバクター・コリの存在を検出する方法であって、該 方法が、 a)請求項6又は7に記載のプライマーの対に前記生物試料を接触させるステ ップであって、前記生物試料中に含まれるカンピロバクター・コリの核酸が、必 要に応じて、カンピロバクター・コリに属する核酸とのプライマーのハイブリダ イゼーションを許容する条件下で、先にハイブリダイゼーションにアクセス可能 にされているステップと、 b)前記カンピロバクター・コリに属する核酸を増幅させるステップと、 c)前記プライマーにより囲まれたフラグメントに相当するカンピロバクター ・コリ の核酸のフラグメントの増幅を証明するステップと、 d)可能であれば、例えば特異的プローブハイブリダイゼーションにより、配 列決定により又は制限部位分析により前記増幅されたフラグメント配列を確認す るステップと、 を含むことを特徴とする方法。 10.生物試料中のカンピロバクター・コリの存在を検出するためのキットであ って、 請求項6又は7に記載の一対のプライマーと、 カンピロバクター・コリの核酸の増幅を行うために必要な試薬と、 可能であれば、増幅されたフラグメントの配列が確認されるのを許容する構成 物、特に請求項4又は5に記載の核酸プローブと、 を含むことを特徴とするキット。
