Description
(式中、R1、R3及びR4は、互いに同一であっても異なっていてもよく、水素原子、炭素数1〜6のアルキル、炭素数1〜6のアルコキシ、炭素数3〜6のシクロアルキル、炭素数6〜20のアリール又は炭素数2〜20のヘテロアリールであり;R2は、水素原子であり;環Arは、置換もしくは無置換の芳香族炭化水素環又は芳香族複素環であり;Aは、−O−、−S−、−Se−又は−NR5−であり、そしてR5は、水素原子、炭素数1〜6のアルキル又は炭素数6〜20のアリールである)
で示される化合物とハロゲン化剤を、ハロゲン間化合物の存在下に反応させることを特徴とする、一般式(2):
(式中、R1、R3、R4、環Ar及びAは、前記と同義であり、Xは、塩素原子又は臭素原子を示す)
で示される化合物の製造方法に関する。
本発明において「炭素数1〜6のアルコキシ」は、基−ORaであって、Raが、上記で定義したとおりの炭素数1〜6のアルキルである基を意味し、例えば、メトキシ、エトキシ、プロポキシ、イソプロポキシ、n−ブチルオキシ、イソブチルオキシ、s−ブチルオキシ、t−ブチルオキシ、ペンチルオキシ又はヘキシルオキシなどが挙げられる。
(式中、R1、R3及びR4は、互いに同一であっても異なっていてもよく、水素原子、炭素数1〜6のアルキル、炭素数1〜6のアルコキシ、炭素数3〜6のシクロアルキル、炭素数6〜20のアリール又は炭素数2〜20のヘテロアリールであり;R2は、水素原子であり;環Arは、置換もしくは無置換の芳香族炭化水素環又は芳香族複素環であり;Aは、−O−、−S−、−Se−又は−NR5−であり、そしてR5は、水素原子、炭素数1〜6のアルキル基又は炭素数6〜20のアリールである)
で示される化合物とハロゲン化剤を、ハロゲン間化合物触媒の存在下、反応させることを特徴とする、一般式(2):
(式中、R1、R3、R4、環Ar及びAは、前記と同義であり、Xは、塩素原子又は臭素原子を示す。)
で示される化合物を得るものである。
(式中、R1、R2、R3、R4、環Ar及びAは、前記と同義であり、R6、R7、R8及びR9は、互いに同一であっても異なっていてもよく、水素原子、炭素数1〜6のアルキル、炭素数1〜6のアルコキシ、炭素数3〜6のシクロアルキル、炭素数6〜20のアリール又は炭素数2〜20のヘテロアリールである)で示される化合物である。その中でも、一般式(1a)において、Aが−O−又は−S−であり、R1、R3、R4、R6、R7、R8及びR9が水素原子又は炭素数1〜6のアルキルであるものが好ましく、Aが−O−又は−S−であり、R1、R3、R4、R6、R7、R8及びR9が全て水素原子であるものがより好ましい。
I−X′ (3)
(式中X′は、塩素原子又は臭素原子を示す)
で表される化合物であり、不純物抑制の観点からX′が臭素原子である化合物がより好ましい。具体的には、一塩化ヨウ素、一臭化ヨウ素などが挙げられ、これらの化合物を単独で又は2種以上混合して用いてもよい。
検出器 :SPD20A(株式会社島津製作所製)
オーブン :CTO−20A(株式会社島津製作所製)
ポンプ :LC−20AD(株式会社島津製作所製)
カラム :ODS−80Tm(東ソー株式会社製)
カラム温度:40℃
溶離液 :アセトニトリル:水=75:25
流速 :1.0ml/min
波長 :254nm
アルゴン雰囲気下、四つ口フラスコに、ジベンゾフラン4.0g(23.8mmol)(東京化成工業株式会社製)、塩化メチレン40gを加え、撹拌下、内温約−50〜−40℃まで冷却した。冷却後、一臭化ヨウ素0.1g(0.5mmol)(Aldrich社製)を添加し、同温度にて、臭素3.8g(23.8mmol)と塩化メチレン7.7gを混合した溶液を滴下し、1時間反応させた。得られた反応液のHPLC純度は、2−ブロモジベンゾフラン82.4%(3−ブロモジベンゾフラン10.5%)であった。
得られた反応液を亜硫酸ナトリウム水溶液で還元し、有機層を分離した。得られた有機層を水洗し、濃縮により溶媒を留去した後、得られた固体残留物にモノクロロベンゼン5ml、イソプロピルアルコール25mlを加えた。次いで、氷水冷にて結晶を析出させた後、濾過を行い、乾燥することでHPLC純度99%の2−ブロモジベンゾフラン4.5gを得た(収率77%、3−ブロモジベンゾフラン非検出)。外観:白色粉末性結晶。
実施例1の一臭化ヨウ素を一塩化ヨウ素0.1g(0.6mmol)(和光純薬工業株式会社製)に変更した以外は、同様の操作を行った。得られた反応液のHPLC純度は、2−ブロモジベンゾフラン79.2%(3−ブロモジベンゾフラン11.6%)であった。
実施例1の一臭化ヨウ素をヨウ素0.06g(0.2mmol)(和光純薬工業株式会社製)に変更し、反応液中で一臭化ヨウ素を調整した以外は同様の操作を行った。得られた反応液のHPLC純度は、2−ブロモジベンゾフラン82.8%(3−ブロモジベンゾフラン10.3%)であった。
実施例1の一臭化ヨウ素をヨウ素0.6g(2.4mmol)(和光純薬工業株式会社製)に変更し、反応液中で一臭化ヨウ素を調整した以外は同様の操作を行った。得られた反応液のHPLC純度は、2−ブロモジベンゾフラン82.9%(3−ブロモジベンゾフラン7.8%)であった。
実施例3のジベンゾフランをジベンゾチオフェン4.0g(21.8mmol)(Aldrich社製)に変更した以外は同様の操作を行った。得られた反応液のHPLC純度は、2−ブロモジベンゾチオフェン90.9%(3−ブロモジベンゾチオフェン1.3%)であった。
四つ口フラスコに、ジベンゾフラン5.0g(29.7mmol)(東京化成工業株式会社製)と酢酸30mlを加え、撹拌下、内温50℃まで昇温した。同温度にて臭素6.3g(39.2mmol)を約20分かけて滴下した。滴下後、室温まで冷却し、1晩反応させた。得られた反応液のHPLC純度は、2−ブロモジベンゾフラン58.7%(3−ブロモジベンゾフラン16.5%)であった。
実施例1の一臭化ヨウ素を塩化鉄(III)0.04g(0.2mmol)(和光純薬工業株式会社製)に変更した以外は同様の操作を行った。得られた反応液のHPLC純度は、2−ブロモジベンゾフラン66.2%(3−ブロモジベンゾフラン17.5%)であった。

- 前記ハロゲン間化合物が、一般式(3):

(式中、X′は、塩素原子又は臭素原子を示す)
で示される化合物である、請求項1に記載の製造方法。 - 前記一般式(3)のX′が臭素原子である、請求項2に記載の製造方法。
- 前記一般式(1)において、環Arが無置換のベンゼンである、請求項1〜3のいずれかに記載の製造方法。
- 前記臭素化剤が、臭素、N−ブロモスクシンイミド又は1,3−ジブロモ−5,5−ジメチルヒダントインである、請求項1に記載の製造方法。
