Claims (19)

  1. 細胞破壊および核酸処理を可能にする条件下で直接的にバイサルファイト薬剤で生体サンプルを処理する工程と;
    処理サンプルからバイサルファイト薬剤を除去する工程と;
    処理サンプル中でターゲット核酸を検出する工程
    を含む、分子検出アッセイ。
  2. 前記生体サンプルは、糞便、鼻水、血液、血漿、血清、頬細胞、膿、創傷、濃縮濾液、脳脊髄液、***、液状化検体細胞診(LBC)組織、FFPE、レーザ捕獲細胞、培養細胞、ペレット化細胞、細菌培養物、細菌コロニー、ウイルス懸濁液、吸引物、気管支洗浄液、痰サンプル、環境サンプル、環境濃縮物、食品、原料、水サンプル、または水濃縮物から選択される、請求項1に記載のアッセイ。
  3. 動物における感染症、遺伝的疾患または遺伝的形質を検出するための、請求項1または請求項2に記載のアッセイ。
  4. 前記感染症は微生物によって引き起こされる、請求項3に記載のアッセイ。
  5. 前記微生物は、細菌、ウイルス、ウイロイド、酵母、菌類、プリオン、寄生虫またはアメーバである、請求項4に記載のアッセイ。
  6. 前記遺伝的疾患は、癌、コピー数変異と関連した疾患、遺伝性疾患、環境誘導疾患、発癌因子への暴露によって引き起こされる疾患、ヌクレオチドリピート長の拡大または縮小によって特徴付けられる疾患である、請求項2に記載のアッセイ。
  7. 前記バイサルファイト薬剤は、ナトリウムバイサルファイトまたはナトリウムメタバイサルファイトである、請求項1から請求項6の何れか一項に記載のアッセイ。
  8. 前記バイサルファイト薬剤は、pH範囲が4.5から5.5で2.5Mから3.5Mの濃度で用いられる、請求項7に記載のアッセイ。
  9. 前記バイサルファイト薬剤は、5.0のpHにて3Mの濃度で用いられる、請求項8に記載のアッセイ。
  10. 前記処理工程は、1分間から30分間、75℃から95℃の間で加熱することを伴う、請求項1から請求項9の何れか一項に記載のアッセイ。
  11. 前記サンプルは、10から20分間、80℃から95℃の間の温度で加熱される、請求項10に記載のアッセイ。
  12. 前記バイサルファイトは、カラムベースの精製、ビーズベースの精製または沈殿によって除去される、請求項1から請求項11の何れか一項に記載のアッセイ。
  13. 前記バイサルファイトの除去後、前記処理サンプルは、pHが少なくとも10である溶出バッファ中に再懸濁される、請求項1から請求項12の何れか一項に記載のアッセイ。
  14. 前記溶出バッファは、pHが11.5から12.5までである、請求項13に記載のアッセイ。
  15. 前記溶出バッファは、pHが少なくとも12である、請求項14に記載のアッセイ。
  16. 核酸プライマーまたはプローブを前記処理サンプルに与える工程をさらに含む、請求項1から請求項15の何れか一項に記載のアッセイ。
  17. 前記ターゲット核酸分子は増幅によって検出される、請求項16に記載のアッセイ。
  18. 前記増幅は、PCR、qPCR、逆転写PCR、デジタルPCR、等温増幅またはシグナル増幅である、請求項17に記載のアッセイ。
  19. 前記ターゲット核酸は、前記処理サンプルを配列決定することによって検出される、請求項1から請求項18の何れか一項に記載のアッセイ。
