Experimental example 1
Research on influence of hypertension fast-decreasing pill on two-kidney one-clamp type hypertension rat
According to clinical indications of the hypertension quick-reducing pill, a rat model with two kidney-clamping type hypertension copied by an operation is selected, and related pharmacodynamic observation indexes are selected according to the hypertension effect to study the treatment effect of the hypertension rat model.
1 materials of the experiment
1.1 Experimental animals
SD rats, male, 10 weeks old, SPF grade, 83, body weight (250 + -20) g, purchased from Tokyo Wintolite laboratory animals technologies, Inc.
1.2 drugs and reagents
The hypertension-treating bolus prepared in example 1; .

2 method of experiment
2.1 two-kidney one-clamp type hypertension rat model establishment
A10-week-old male SD rat is fed adaptively for 1 week, then blood pressure basic value measurement is carried out, under a quiet environment, a conscious rat is placed into a rat intelligent noninvasive blood pressure measuring instrument fixing belt to be preheated for 5-10 min, after pulse waves are stabilized, tail arterial systolic pressure (SBP) is measured continuously for 3 times, and an average value is taken as an SBP basic value. Except 10 rats were used as normal controls (blank group), other rats underwent left renal aortic stenosis surgery (two kidneys with one clip) to replicate the hypertension model.
Stopping feeding 12h before operation and freely drinking water; metal instruments, glass instruments, silk threads and the like for operation are subjected to high-pressure sterilization before operation. During operation, 40mg/kg of sodium pentobarbital is used for carrying out intraperitoneal injection and anaesthetizing rats, an operating table is fixed on the supine position, the furs of about 3cm multiplied by 4cm in the left abdominal area and the left kidney area are cut off, 1% iodine tincture and 75% ethanol are respectively used for disinfecting the skin, the skin is cut by about 3cm, the cut is parallel to the abdominal median line, the skin is fixed by a skin pulling hook, an abdominal muscle layer is separated bluntly by hemostatic forceps, the surgical forceps carefully push away the intestinal canal, pancreas and other internal organs to expose the left kidney, the renal artery is carefully separated by a glass needle, the left renal artery of the rat is clamped by a U-shaped silver clip of 0.25mm, the muscle and the skin are sequentially sutured, the normal group of rats is not treated, and the rat is continuously fed with 3d ip penicillin injection (10 ten. Blood pressure monitoring is carried out continuously for four weeks every week after operation, the blood pressure value at the 4 th week is referred, SBP is more than or equal to 160mmHg before and after model making is compared, and the model replication success is determined by the increase of 20 mmHg.
2.2 grouping and administration
The rats with successful model taking are randomly divided into 4 groups according to the SBP value, namely a model group, a positive drug captopril group, a hypertension quick-reducing pill medium dose group and a hypertension quick-reducing pill low dose group, the number of the mice is shown in table 2, the rats are continuously administrated for 4 weeks, the blood pressure value is monitored once per week, and the drug dose is adjusted according to the change condition of the SBP.
Wherein the dosage of the hypertension quick-reducing pill is equivalent to the equivalent dosage of the clinical dosage of human; the low dose of the hypertension pill is 1/2 which is equivalent to the dose of human clinical medication, and the specific administration condition is shown in Table 1.

2.3 detection of animal index
Monitoring blood pressure once a week after administration, taking materials after four weeks, fasting for 12h before material taking, weighing fasting body mass of all animals, after 1h of last administration, injecting 40mg/kg sodium pentobarbital into abdominal cavity for anesthesia, taking blood from abdominal aorta, anticoagulating heparin, and separating plasma for measuring Aldosterone (ALD) and angiotensin (Ang II).
2.4 statistical methods
Statistical analysis was performed using a sps 17.0 and data was recorded using "± s". The data fit to a normal distribution, multiple sample mean comparisons were performed using one-way-ANOVA, with all subjects tested with LSD and all subjects tested with Dunnett's T3, and P <0.05 was statistically significant.
3 results of the experiment
3.1 Effect of hypertension-treating pill on blood pressure of hypertensive rat with two kidneys and one clip
The experimental result shows that the basal SBP of rats in each group has no significant difference (p is more than 0.05) compared with each other;
after four weeks of two-kidney one-clamp surgery, the SBP appeared significantly higher in each surgery group compared to the blank group (mean P <0.01), indicating successful model replication.
After four weeks of administration, the model group and rats in each administration group had significantly increased SBP (mean P <0.01) compared to the blank group; however, the blood pressure values of the captopril group and the hypertension pills in the middle and low dose groups were significantly decreased (P <0.01, 0.05, 0.01, respectively) compared to the model group, and the results are shown in table 2.

3.2 Effect of hypertension-lowering pill on angiotensin and aldosterone in blood plasma of hypertensive rat with two kidneys and one clip
The experimental result shows that compared with the blank group, the angiotensin in the plasma of the model group is obviously increased and has significant difference (P < 0.01). Compared with the model group, the positive medicine captopril, the high blood pressure quick-reducing pill and the low dosage can obviously reduce the content of angiotensin in blood plasma, and have obvious difference (P <0.01, P <0.01, P < 0.05).
The plasma aldosterone content of the model group tended to increase compared to the blank group; compared with a model group, the dosage group in the hypertension quick-reducing pill can reduce the content of aldosterone in blood plasma, and the trends of the dosage group have certain physiological significance, but no statistical difference occurs; as described in table 3.

4 discussion of Experimental results
The two-kidney one-clamp type hypertension animal model is a common experimental model for evaluating the antihypertensive effect of drugs, and the mechanism of the hypertension model is renal ischemia caused by renal artery constriction, renin-angiotensin-aldosterone system (RAAS) activation induced by renin-angiotensin-aldosterone system increase in kidney and secondary. The experimental result shows that stable hypertension is formed in rats 4 weeks after two-kidney one-clamp operation, and the significant increase of Ang II in plasma indicates that the model is successfully copied and accords with the literature report. The hypertensive fast-reducing pill can obviously reduce the tail arterial systolic pressure of a model rat on the basis of successful model replication after being continuously administrated for 4 weeks, reduce the obvious expression of Ang II caused by operation in plasma, and show the trend of reducing the content of the high-expression aldosterone, which indicates that the hypertensive fast-reducing pill has better antihypertensive effect on two-kidney one-clamp type hypertensive rat, and the mechanism is realized by regulating RAAS system.
It should be understood that the above examples and experimental examples are only examples for clearly illustrating the present invention, and are not intended to limit the present invention.
