Abstract
本发明公开了一种降血糖的纳米包合物及其制备方法,属于功能食品技术领域。本发明通过酸溶再加入碱液中和的方式,将灰树花提取物、膳食纤维、蛋白肽制备成纳米包合物。本发明的纳米包合物原料安全、配伍合理。并且该包合物的制备方法与工艺,操作简易、重现性良好,所得颗粒或压片外形美观、耐磨性强、成形性好。该包合物经实验验证,具有耐胃酸、缓释的效果,且在高血糖小鼠模型中,灌胃给药2周后,模型的空腹血糖下降率为39.5%,可有效改善胰岛β细胞功能,降低血糖。该组方口感优良,可作为固体饮料或功能食品原料进行应用,广泛适用于糖尿病患者。
Description
技术领域
本发明涉及一种降血糖的纳米包合物及其制备方法,属于功能食品技术领域。
背景技术
糖尿病是一种由于胰岛素分泌缺陷或胰岛素作用障碍所致的以高血糖为特征的代谢性疾病。持续高血糖与长期代谢紊乱等可导致全身组织器官,特别是眼、肾、心血管及神经***的损害及其功能障碍和衰竭。严重者可引起失水,电解质紊乱和酸碱平衡失调等急性并发症酮症酸中毒和高渗昏迷。随着人们生活条件的不断改善与提高,物质生活的丰富多样化,加之快节奏的生活,使得糖尿病等多发病症患者人数急剧增加。
现代医学对糖尿病的控制主要从治疗角度出发,针对性地采用口服或注射降糖药、胰岛素等方式,但这些药物极易引起患者低血糖和胰岛β细胞功能衰竭及其他副作用,且难以完全杜绝或者防止慢性并发症的出现,尤其是某些糖尿病的特异性慢性并发症。饮食对糖尿病病程影响深远,合理补充具有降糖作用的食品不仅有利于糖尿病病情的控制,同时有利于病人机体的营养补给。因此,寻找开发安全、有效的降血糖功能食品是目前研究的热点之一。
灰树花Boletus frondosus Dicks,多孔菌科真菌贝叶多孔菌的子实体,作为一种珍稀的药食兼用菌,其富含数百种活性多糖、甾类、酚类、三萜类等活性物质。研究表明,从人工培养的灰树花子实体粉经乙醇***回流、水提、醇沉、干燥得粗多糖能够有效抑制α-糖苷酶活性,降低高血糖小鼠的血糖水平。但是文献研究中多用***等有机溶剂进行提取,残留溶剂难以去除,不适用于食品开发。目前常见的灰树花功能产品主要以口服溶液或压片糖果为主,而其含有的主要功效成分-多糖存在胃液易被酸降解,分子量大导致吸收差等问题,因此也有研究者将其制备成纳米乳等以提高生物利用度,但纳米乳配方组成、制备工艺较为复杂。鉴于以上,本发明旨在发明一种工艺简单、生物利用度高的灰树花功能食品。
发明内容
为解决上述技术问题,本发明提供一种降血糖的纳米包合物,具有耐胃酸、缓释的效果,且制备工艺简单,重复性好,本发明的包合物主要功能原料为灰树花提取物、膳食纤维和蛋白质。针对糖尿病人三多一少症状,提供功能性灰树花提取物并辅以合理营养,以缓减糖尿病人临床营养情况以及症状。
本发明的第一个目的是提供一种降血糖的纳米包合物的制备方法,包括如下步骤:将水不溶性膳食纤维溶于酸溶液中,加入灰树花提取物、水溶性膳食纤维和蛋白肽搅拌,混合均匀后,在体系中加入碱液中和,干燥粉碎得到所述的降血糖的纳米包合物。
进一步地,所述的纳米包合物中灰树花提取物的质量分数不低于10%。
进一步地,所述的灰树花提取物、水溶性膳食纤维、水不溶性膳食纤维、蛋白肽的质量比为10~35:4~6:4~6:1~5。
进一步地,所述的水溶性膳食纤维为魔芋葡甘聚糖,水不溶性膳食纤维为壳聚糖或壳寡糖。
进一步地,所述的蛋白肽为植物蛋白来源蛋白肽或动物蛋白来源蛋白肽。
进一步地,所述的蛋白肽为大豆蛋白肽、玉米蛋白肽、花生蛋白肽、胶原蛋白肽、乳清蛋白肽中的一种或一种以上组合。
进一步地,所述的灰树花提取物按照如下方法制备得到:取灰树花粉末,加入水:醇体积比为2~4:1的醇溶液进行回流提取,干燥得到灰树花提取物。
进一步地,所述的酸溶液为冰醋酸、盐酸中的一种或一种以上组合。
进一步地,所述的碱液为氢氧化钠、氢氧化钾、氢氧化钙、碳酸钠、碳酸钾、碳酸氢钠中的一种或一种以上组合。
进一步地,碱加入量按化学计量计算为最终溶液呈pH=6.5~7.5。
进一步地,所述的制备方法还包括将粉碎后的纳米包合物与辅料混合,制备成颗粒或压片。
进一步地,所述的辅料包括甜菊糖苷0.1~1份、天然香精0.01~0.1份。
本发明的第二个目的是提供所述方法制备得到的降血糖的纳米包合物。
本发明的第三个目的是提供包含所述的降血糖的纳米包合物的食品、药品或保健品。
本发明的有益效果是:
本发明提供了一种具有降血糖功能的纳米包合物,该纳米包合物原料安全、配伍合理。并且该包合物的制备方法与工艺,操作简易、重现性良好,所得颗粒或压片外形美观、耐磨性强、成形性好。该包合物经实验验证,具有耐胃酸、缓释的效果,且在高血糖小鼠模型中,灌胃给药2周后,模型的空腹血糖下降率为39.5%,可有效改善胰岛β细胞功能,降低血糖。该组方口感优良,可作为固体饮料或功能食品原料进行应用,广泛适用于糖尿病患者。

具体实施方式
下面结合附图和具体实施例对本发明作进一步说明,以使本领域的技术人员可以更好地理解本发明并能予以实施,但所举实施例不作为对本发明的限定。
实施例1:灰树花原料提取
取人工培养的灰树花子实体粉末10份,加入30份水:乙醇=3:1(体积比)的乙醇水溶液进行回流提取2h,干燥得到提取物。
实施例2:包合物制备工艺
取壳聚糖5份,溶于500mL 0.1%醋酸溶液中,加入灰树花提取物35份、魔芋葡甘聚糖5份、胶原蛋白肽1份,搅拌混合均匀,并在搅拌状态下加入1%碳酸氢钠溶液,直至pH=7,冷冻干燥30h,置于透射电镜下观察,粒径约为20±5nm,制粒、烘干、压片,制成1g/片的片剂。
实施例3:包合物压片制备工艺
取壳聚糖5份,溶于500mL 0.1%醋酸溶液中,加入灰树花提取物20份、魔芋葡甘聚糖5份、胶原蛋白肽1份,搅拌混合均匀,并在搅拌状态下加入1%碳酸氢钠溶液直至pH=7,冷冻干燥30小时,粉碎后,与甜菊糖苷0.1份、天然香精0.05份混匀,制粒、烘干、压片,制成1g/片的片剂。所得压片光泽明显,口感良好,气味宜人。
实施例4:包合物制备
取壳寡糖5份,溶于500mL 0.1%醋酸溶液中,加入灰树花提取物20份、魔芋葡甘聚糖5份、大豆蛋白肽1份,搅拌混合均匀,并在搅拌状态下加入1%碳酸氢钠溶液,直至pH=7,冷冻干燥30小时,制粒、烘干、压片,制成1g/片的片剂。
实施例5:包合物制备
取壳聚糖5份,溶于500mL0.1%盐酸溶液中,加入灰树花提取物20份、魔芋葡甘聚糖5份、大豆蛋白肽5份,搅拌混合均匀,并在搅拌状态下加入1%碳酸氢钠溶液,直至pH=7,冷冻干燥30小时,制粒、烘干、压片,制成1g/片的片剂。
对比例1:
取灰树花提取物35份、壳寡糖5份、魔芋葡甘聚糖5份、胶原蛋白肽1份,搅拌混合均匀,制粒、烘干、压片,制成1g/片的片剂。
对比例2:
取灰树花提取物35份、壳寡糖5份、魔芋葡甘聚糖5份、胶原蛋白肽1份,并加入甜菊糖苷0.1份、天然香精0.05份,搅拌混合均匀,制粒、烘干、压片,制成1g/片的片剂。
实施例6:胃液稳定性评价
在模拟人工胃液中测试不同方法制备所得的产品的胃液稳定性能。
人工胃液配方:取质量分数为0.1mol/L的盐酸1.0mL,用蒸馏水稀释将其pH值调至2.0。然后加入胃蛋白酶0.01g/mL,溶解均匀后用微孔滤膜除菌备用。
将实施例1-4以及对比例1-2制得的产品分别取1片加入到上述人工体外模拟胃液中,每1h取一次样,采用考马斯亮蓝法检测样品中蛋白浓度,以在595nm处的OD表示,每个样品重复测试3次。由表1数据可知,几种样品在初始时溶液的OD值相近,有一定的本底吸收。随时间推移,对比例1与对比例2制得的压片在30min内完全崩解。而实施例1的产品在胃排空时间(4h)内只释放了(8.0±0.32)%。

实施例7:包合物降糖效果评价
取实施例3制备得到的包合物压片,建立高血糖动物模型,分成模型组和包合物组,模型组灌胃生理盐水,包合物组灌胃包合物混悬液,连续灌胃给药14天,每天监测小鼠体重、饮水量和饮食量、血糖值。结果表明,包合物组小鼠糖尿病的三多一少症状显著改善,且降糖效果显著。
另按照文献方法:取人工培养的灰树花子实体粉末,加入乙醇:***=3:1(体积比)的乙醇***溶液进行回流提取,然后加入水进行水提,再使用乙醇进行醇沉,干燥得到提取物。然后按照实施例3的方法制备包合物压片,进行降糖效果评价,与实施例3方法得到的包合物压片降糖效果相当。
以上所述实施例仅是为充分说明本发明而所举的较佳的实施例,本发明的保护范围不限于此。本技术领域的技术人员在本发明基础上所作的等同替代或变换,均在本发明的保护范围之内。本发明的保护范围以权利要求书为准。
Claims (10)
1.一种降血糖的纳米包合物的制备方法,其特征在于,包括如下步骤:将水不溶性膳食纤维溶于酸溶液中,加入灰树花提取物、水溶性膳食纤维和蛋白肽搅拌,混合均匀后,在体系中加入碱液中和,干燥粉碎得到所述的降血糖的纳米包合物。
2.根据权利要求1所述的方法,其特征在于,所述的纳米包合物中灰树花提取物的质量分数不低于10%。
3.根据权利要求1所述的方法,其特征在于,所述的灰树花提取物、水溶性膳食纤维、水不溶性膳食纤维、蛋白肽的质量比为10~35:4~6:4~6:1~5。
4.根据权利要求1所述的方法,其特征在于,所述的水溶性膳食纤维为魔芋葡甘聚糖,水不溶性膳食纤维为壳聚糖或壳寡糖。
5.根据权利要求1所述的方法,其特征在于,所述的蛋白肽为大豆蛋白肽、玉米蛋白肽、花生蛋白肽、胶原蛋白肽、乳清蛋白肽中的一种或一种以上组合。
6.根据权利要求1所述的方法,其特征在于,所述的灰树花提取物按照如下方法制备得到:取灰树花粉末,加入水:醇体积比为2~4:1的醇溶液进行回流提取,干燥得到灰树花提取物。
7.根据权利要求1所述的方法,其特征在于,碱液加入量按化学计量计算为最终溶液呈pH=6.5~7.5。
8.根据权利要求1所述的方法,其特征在于,所述的制备方法还包括将粉碎后的纳米包合物与辅料混合,制备成颗粒或压片。
9.一种权利要求1-8任一项所述的方法制备得到的降血糖的纳米包合物。
10.一种包含权利要求9所述的降血糖的纳米包合物的食品、药品或保健品。
